ObjectiveTo detect the serum concentration of TNF-α,sTNFR1, sTNFR2and theperipheral blood levels of CD4+CD25highregulatory T cells in Parkinson’sdisease. To investigate the reasens and related factords of those changes andmechanism of the immune inflammatory response in Parkinson’s disease.MethodsThe serum concentration of TNF-α,sTNFR1, sTNFR2in40PD patients and30healthy subjects with age and sex-matched were measured byEnzyme-Linked Immunolsorbent Assay(ELISA). And the percentage ofcirculating CD4+CD25+regulatory T cells and CD4+CD25highregulatory T cellswere detected by using flow cytometry.The United Parkinson’s Disease RateScale Part III Score(UPDRS III)and Modified Hoehn and Yahr Staging Scale(HY) were evaluated in all studied PD patients.And all PD patients were dividedinto Several subgroups according to the treatment status.ResultsThe mean serum sTNFR1levels are significantly higher and thepercentage of circulating CD4+CD25+regulatory T cells and CD4+CD25highregulatory T cells are significantly lower than healthy subjects group(P<0.05,P<0.01).there was a low positive correlation between the levels of TNF-α and sTNFR1in PD group.(rs=0.33,P<0.05),there was a significantpositive correlation between the levels of sTNFR1and sTNFR2in PDgroup(rs=0.58,P<0.01),there was no correlation TNF-α and sTNFR2(rs=0.26,P>0.05).there was a moderate positive correlation between the levels ofsTNFR1and sTNFR2level in healthy group(rs=0.42, P <0.05)there was nosignificant correlation TNF-α and age,duration,UPDRS III score,Hoehn-YahrStaging Scale in PD group,there was no significant correlation betweensTNFR1,sTNFR2and duration,UPDRS III score,Hoehn-Yahr Staging Scale inPD group,there was a significant positive correlation between sTNFR1and agein PD group(rs=0.521,P=0.001),there was a low positive correlation betweensTNFR2and age in PD group (rs=0.35,P<0.05);The PD group was dividedinto different the subgroups according to the diffierent state of therapies ofillness,there is no significant difference(allP>0.05).ConclusionsThe mean serum sTNFR1levels were significantly higher and thepercentage of circulating CD4+CD25+regulatory T cells and CD4+CD25highregulatory T cells were significantly lower in PD group.Our fingdings suggestthat all those immunity factors were detected in our research may implicate inPD pathogenesis.The percentage of circulating CD4+CD25highregulatory T cellswere significantly lower and sTNFR1were significantly higher in PD,Whichsuggest that the immune status of abnormal with PD patients.The mean serumsTNFR1levels were significantly higher,there was a significant positivecorrelation between sTNFR1and age in PD group,there was a low positivecorrelation between sTNFR2and age in PD group.Our finding suggest thatTNF-α mediated intracellular signaling cascades involved in immuneinflammatory pathological process of PD,aging is the major risk factors.Therewas a low positive correlation between the levels of TNF-α and sTNFR1in PDgroup.So measurement of circulating levels of the two sTNFRs is useful to determine the overall production of TNF-α,which may be considered morereliable markers of inflammatory activity than TNF-α.
