Objective: To investigate the expression and correlation of breast cancermetastasis-suppressor1(BRMS1)and special AT rich sequence binding protein1(SATB1) in human laryngeal squamous cell carcinoma and the adjacent tissues ofcancer, this study was aimed to explore the relationship and the clinical significanceof these two genes.Methods:1. Examine the expression of BRMS1and SATB1in both20cases of supraglotticlarynx squamous cell carcinoma and20cases of glottic carcinoma,as well as examinethe expression of two genes in corresponding adjacent normal tissues of cancer byusing immunohistochemistry EliVision technique.2.Detect the expression of BRMS1mRNA and SATB1mRNA in both20cases ofsupraglottic larynx squamous cell carcinoma and20cases of glottic carcinoma,as wellas examine the expression of two genes in corresponding adjacent normal tissues ofcancer by reverse transcription polymerase chain reaction (RT-PCR).3.To analyze the relationship among the expression of BRMS1、SATB1in the age,gender, TNM stages, grades of differentiation,metastasis of lymph nodes and therelativity of their expression in laryngeal squamous cell carcinoma.Results:一.The results detected by IHC1.In supraglottic larynx squamous cell carcinoma,the positive rate of BRMS1proteinis lower than the positive rate in corresponding adjacent normal tissues of cancer, theresult was significantly different between two groups(P<0.05).2. In glottic larynx squamous cell carcinoma,the positive rate of BRMS1protein islower than the positive rate in corresponding adjacent normal tissues of cancer, theresult was significantly different between two groups(P<0.05).3.There is no significantly different of the positive rate of BRMS1protein between in supraglottic larynx squamous cell carcinoma and in glottic larynx squamous cellcarcinoma(P>0.05).4.In supraglottic larynx squamous cell carcinoma,the positive rate of SATB1proteinis higher than the positive rate in corresponding adjacent normal tissues of cancer,the result was significantly different between two groups(P<0.05).5.In glottic larynx squamous cell carcinoma,the positive rate of SATB1protein ishigher than the positive rate in corresponding adjacent normal tissues of cancer, theresult was significantly different between two groups (P<0.05)6.There is no significantly different of the positive rate of SATB1protein between insupraglottic larynx squamous cell carcinoma and in glottic larynx squamous cellcarcinoma(P>0.05).7.In supraglottic larynx squamous cell carcinoma,the expression of BRMS1andSATB1protein are related to TNM stages, grades of differentiation,metastasis oflymph nodes,but are not related to age,gender.8..In glottic larynx squamous cell carcinoma,the expression of BRMS1and SATB1protein are related to TNM stages, grades of differentiation,but are not related toage,gender,metastasis of lymph nodes.9.No matter in supraglottic or glottic larynx squamous cell carcinoma, the expression ofBRMS1protein was negatively correlated to SATB1protein.二、The results detected by RT-PCR1.Both in supraglottic larynx squamous cell carcinoma and glottic larynx squamouscell carcinoma,the expression of BRMS1mRNA is lower than the expression incorresponding adjacent normal tissues of cancer, the result was significantly differentbetween two groups(P<0.05).2.Both in supraglottic larynx squamous cell carcinoma and glottic larynx squamouscell carcinoma,the expression of SATB1mRNA is higher than the expression incorresponding adjacent normal tissues of cancer, the result was significantly differentbetween two groups(P<0.05).Conclusion:1.No matter in supraglottic or glottic larynx squamous cell carcinoma,the expression of BRMS1is lower than the expression in corresponding adjacent normal tissues ofcancer.2. No matter in supraglottic or glottic larynx squamous cell carcinoma,the expressionof BRMS1is higher than the expression in corresponding adjacent normal tissues ofcancer.3.In supraglottic larynx squamous cell carcinoma,the expression of BRMS1andSATB1are related to TNM stages, grades of differentiation,metastasis of lymphnodes,but are not related to age,gender.4.In glottic larynx squamous cell carcinoma,the expression of BRMS1and SATB1are related to TNM stages, grades of differentiation,but are not related toage,gender,metastasis of lymph nodes.5. The expression of BRMS1was negatively correlated to SATB1,combined analyses of theexpression of BRMS1and SATB1might be the important index of the lymphaticmetastasis in laryngeal squamous cell carcinoma.
