| Objective:This experiment was to observe the effects of Yishenjiangzhuo Prescription on the serum SOD〠MDA〠TNF-a of5/6nephrectomy CRF rats by using the method of animal experiments and to explore the efficacy mechanism of Yishenjiangzhuo Prescription in the treatment of CRF.The will provide a experimental evidence for the use of Yishenjiangzhuo Prescription.Method:60male SD rats were randomly divided into five groups:normal group12, sham group12,model group12,Niaoduqing control group12Yishenjiangzhuo Prescription treatment group12.Model group, Niaoduqing control group, Yishenjiangzhuo Prescription treatment group for5/6nephrectomy surgery to establish CRF rats mode; The normal group without surgery, performed the sham group with sham surgery; Four weeks after surgery, in the daily morning, the control group, the treatment group was gavaged56days respectively with Niaoduqing particles,Yishenjiangzhuo Prescription; normal group, sham operation group, model group were given saline.The rats at7PM of the56th day morning are placed in the metabolic cage to collect24hour urine sample and record the amount of urine at7:00the next day.The urine sample will be detected urinary β2-MG, urinary NAG, urinary RBP. The blood sample collected by abdominal aortic is used to detected the levels of RBC, Hb and serum Scr, BUN, SOD, MDA, TNF-a, β2-MG in groups of rats. Taking the kidney tissue is to observe pathological morphology.Result:①Compared with the control group, model group RBC, Hb, SOD were significantly decreased, the levels of serum Scr, BUN, MDA, TNF-a, β2-MG and urinary (32-MG, NAG, RBP are significantly increased, the difference is significant (P<0.01).②Compared with the model group, Niaoduqing particle group and Yishenjiangzhuo Decoction group significantly increase RBC, Hb, SOD levels, decreasing serum Scr, BUN, MDA, TNF-a, β2-MG and urinary β2-MG, NAG, RBP levels, the difference is significant (P<0.01).③Compared with Niaoduqing particle group,Yishenjiangzhuo Decoction could significantly elevate RBC, Hb, SOD levels, reducing serum Scr, BUN, MDA, TNF-a, p2-MG and urinary β32-MG, RBP levels, the difference is significant (P<0.01).④Kidney tissue pathomorphology:Under light microscopyg:lomerular, tubular and renal interstitium of the control group and the sham group have no significant change and have normal distribution and arrangement. The sham group occasionally has inflammatory cell infiltration in renal interstitium.;Model group has increased glomerular volume, tubular atrophy, renal tubular epithelial cells edema, interstitial fibrosis, inflammatory cell infiltration in renal interstitium, focal disordered distribution and arrangement;In the control group, there are glomerular edema, tubular atrophy, interstitial fibrosis, yet normal distribution and arrangement;Treatment group has increased glomerular volume, renal tubular epithelial cell edema, interstitial fibrosis, and arrange distribution is still normal.Conclusion:(1)Yishenjiangzhuo Prescription could improve anemia status of CRF rats in certain extent, significantly increase serum SOD of CRF rats, decrease serum MDA, TNF-a levels;(2)Yishenjiangzhuo Prescription significantly reduces serum Scr,BUN of CRF rats to improve renal function in CRF rats;(3)Yishenjiangzhuo Prescription significantly decreases urinary β2-MG, NAG, RBP levels of CRF rats, with improving tubular function of CRF rats;(4)Yishenjiangzhuo Prescription can improve the pathological changes of renal tissue, improve renal interstitial fibrosis an of CRF rats. |
