Objective:To construct a ovary cancer cell line SKOV3with a stableknockdown of RhoC gene by lentiviral vectors, and to observe theinbibitional effect of RhoC gene in ovary cancer cell SKOV3.Methods:Reconstrcted lentivirus with GFP producing RhoC miRNA wastransfected into SKOV3cells,and the stably transfected cells wereobtained after being screened with puromycin. The RhoC mRNA weredetected by real time quantitative PCR.Results:The lentiviral vector was constructed successfully and a stable RhoCknockdown SKOV3cell line was established.The RhoC gene expressionwas efficiently suppressed compared with the control group.Conclusion:The lentivirus-mediated RhoC gene silence can suppress theexpression of RhoC mRNA.... |
