Higher fungi belong to the "creative" natural resource, which contains manysecondary metabolites. Among of this, a single component which has novel structureand biological activity has important significance to the development of modern drugs.In this paper, two higher fungi (Chroogomphis rutilus, Suillus luteus) ethyl acetateextract (CREA, SLEA) was tested as test materials using chromatography, nuclearmagnetic resonance, mass spectrometry and other techniques to analyze thecomposition of the main chemical components. In addition, some of these isolatedproducts, made a preliminary study of antioxidant, anti-tumor activity, the mainresults were listed as below:1. Seven compounds were isolated from the CREA, through NMR and othermethods, they were identified as:(2-1-1) cis, cis-9,12-eighteen two acid, namelylinoleic acid;(2-1-2) cis-9-octadecenoic acid, namely oleic acid;(2-1-3)(22E,24R)-ergostan-5,7,22-trien-3β-alcohol, namely ergosterol;(2-1-4) ergosta-7,22-dien-3-one;(2-1-5) ergosta-4,6,8(14),22-tetraen-3-one (2-1-6)2-amino-3-formyl-6-methoxypyridine;(2-1-7) Polyozellin. Other components’ weightwere too little to test spectrum or impure by NMR detection.2. Eight compounds were isolated from the SLEA, through NMR and othermethods they were identified as:(2-2-1)3,6-dihydroxy-((2E,6E,10E)-3,7,11,15-hexamethyl sixteen carbon-2,6,10,14-tetraene) ethyl benzene;(2-2-2)p-hydroxybenzeneethanol;(2-2-3) dibutyl phthalate;(2-2-4) diisobutyl phthalate;(2-2-5)3β,5α,6β-trihydroxy ergosterol-7,22-diene;(2-2-6) ergosterol peroxide;(2-2-7)4-hydroxyphenethyl acetate;(2-2-8)1-O-β-D-glucopyranosyl-(2S,3R,4E,8E,2’R)-2-N-(2’-hydroxypalmitoyl)-9-methyl-4,8-sphingadienine. Except to compounds2-2-1,2-2-5, other compounds were first isolated from the mushroom Suillus luteus.3. All the fifteen compounds from CERA and SLEA are tested about theantioxidant and antitumor activity. DPPH· scavenging assay was used to evaluate theantioxidant activity. Anti-tumor activity (human gastric cancer BGC-823cells) was measured by MTT assay. The result indicated that most compounds showedantioxidant and antitumor activity in various degrees. Compounds2-1-7,2-2-1revealed the higher capacity on scavenging DPPH free radical with EC50value of0.35mg/Ml and0.0044mg/mL, showed strong antioxidant activity. Meanwhilecompounds2-1-7,2-2-1could strongly inhibite the proliferation of BGC cell line withIC50value51.2μg/mL and7.47μg/mL, showed high anti-tumor ractivity. In addition,the compounds2-1-3,2-2-3,2-2-4,2-2-6have good antioxidant activity, othercompounds do not have significantly biological activity.
