Thyroid carcinoma is the most common malignant tumor in theendocrine system and the morbidity had grown rapidly over the world inrecent years, among which papillary thyroid carcinoma (PTC) is the mostcommon pathological pattern, roughly account for75%-85%of thetotal.Papillary thyroid carcinoma is kind of complicated and alsopolygenic disease still with uncertainty of the existence of major gene forthe carcinoma of thyroid and its pathogenesis has yet to be fullyunderstood.Studies have verified that papillary thyroid carcinoma presents stronggenic factor and it is kind of disease of multifactorial inheritance resultingfrom the combined actions of external environment and polygenes.Papillary thyroid carcinoma is charactered with a complexed geneticbackground. Its genetic susceptibility is likely to be determined bymulti-susceptibility gene with various genepenetrance and it has verylarge heterogeneity of the hereditary phenotypic and its genetic model isdifferent from monogenic disease that is not conformed with Mendel ’ slaw of inheritance. Besides, it has proved that the external environmentplays a critical role in the occurrence and development of papillarythyroid carcinoma. It is a significant research subject trying to recognise and colone the susceptibility gene of thyroid carcinoma. Meanwhile, it isalso a very important subject to create analysis method and researchstrategy for locating carcinoma of thyroid, the kind of multiple-factorinheritary disease at the current phase of research.The geneticsusceptibility of papillary thyroid carcinoma is, at present, regarded as astorage effect generated from heritable variation of multi-loca, whilesingle loca has less effect on genic phenotype.The mode of inheritance of genetic susceptibility is probably a crucialfactor for the occurrence and development of papillary thyroidcarcinoma.The studies have manifested that there are3to20genes jointlycausing every single polygenic disease and the single nucleotidepolymorphism (SNP) with its specific composition of haplotype is theprobable major cause of the susceptibility to the disease.For furtherexploring and revealing the pathogenesis of polygenic disease, it is ofgreat significance of carrying out correlation analysis on candidate geneswhich affect the polygenic disease related to regulatory passage or onsingle nucleotide polymorphism (SNP) of the genetic gene defined bygenome. The studies, domestic as well as abroad, show that there arecorrelations between the single nucleotide polymorphism (SNP) and thesusceptibility of the papillary thyroid carcinoma and the studies on thissubject is meaningful in guiding the early diagnosis and risk assessmentof the papillary thyroid carcinoma. ObjectiveTo investigate the relationship between BRAF gene polymorphismsand papillary thyroid carcinoma susceptibilityMethods:A hospital-based case-control study designed and analysised therelationship between papillary thyroid carcinoma and BRAF genepolymorphism.Results:Differences irradiation history of papillary thyroid cancer patientsbetween different sexes show a statistically significant difference.(χ2=18.218, P<0.05)rs11762469allele and genotype frequencies was not statisticallysignificant difference in the case group and the control group (χ2=0.760P>0.05; χ2=1.392P>0.05); rs1267636allele and genotype frequenciesin the case group and the control group, the difference was notstatistically significant (χ2=0.951P>0.05; χ2=1.069P>0.05);rs17623204allele and genotype frequencies in the case group and thecontrol group, the difference was not statistically significance (χ2=0.008,P>0.05; χ2=0.257P>0.05); rs3748093allele and genotype frequencieswas not statistically significant difference in the case group and thecontrol group (χ2=0.109, P>0.05; χ2=1.397P>0.05)When the results were further analysised by the traditional risk factors for PTC, we found that there was interaction between thers1267636TT genotype and the risk factors for PTC(smoking、drinking)(χ2=12.487, P<0.05;χ2=7.210, P<0.05)ConclusionsBRAF gene’s rs11762469, rs1267636, rs17623204and rs3748093alleles and genotypes may not be related with papillary thyroidcarcinoma.There may be some interactions between the rs1267636TT genotypeand the risk factors of PTC (smoking、drinking).
