Objective:To study the Anti-diabetic effect of an active components group from Momordica Chanrantia in Type2Diabetic Model Mice, determines the main active components of Momordica Chanrantia,and preliminary study on its mechanism of action of lowing blood, providing scientific theory basis for the clinical application of Momordica Chanrantia.Methods:Continuous feeding high fat diet (lard10%,egg yolks10%, bile salts0.2%, cholesterol1%, commercial diet78.8%) for four weeks to establish hyperlipidemia mouse model. And then through intraperitoneal injection of streptozotocin (STZ) to establish mouse model of type II diabetes. Unripe Fresh green fruit of MC purchased from a local market was washed thoroughly with water, cut open,dried and the seeds removed. Twenty five kilograms of Momordica charantia were boiled in30L distilled water for3h, and this extraction process was repeated for3times. The four extracts were combined and concentrated on a rotary evaporator under reduced pressure followed by drying in a freeze dryer. Adding with alcohol volume of95%ethanol in concentrated liquid to the final containing alcohol volume of65%, placing in4℃of refrigerator for24hours, then filtered. Eight fractions of dry extract of Momordica Chanrantia were obtained by double evaporate water,20%methanol,40%methanol,60%methanol,80%methanol and100%methanol.The extract of100%methanol were combined with80%methanol, because they were too weak parts. The extracts of Momordica Chanrantia were rotary evaporated under room temperature followed by drying in a freeze-dryer. The II diabetes mice were randomly assinged to the model group,the metformin group, the group treated with Metformin hydrochloride following extraction with double evaporate water(Fra1),20%methanol(Fra2),40%methanol(Fra3),60%methanol(Fra4),80%methanol and100%methanol (Fra5). Continuously intragastric administration for4weeks, body weight were recorded weekly.4weeks of the treatment later, the body weight, the1evels of fasting blood glucose concentration(GLU), glucose tolerance, total cholesterol (TC), triglyceride (TG), alanine aminotransferase (ALT), aspertate aminotransferase (AST) and other blood biochemical indexes were determined.The activity of the extracts of Momordica Chanrantia has been determined in accordance with the relevant biochemical parameters. At the same time, the changes of related genes including CYP7A1, G6pc and Gck were determined by the way of molecular biology.Lastly, the data were statistically analyzed.Results:1.The different kinds of Momordica Chanrantia extract have good effects on body weight, water intake, food intake and mental state in diabetic mice.2. The different kinds of Momordica Chanrantia extract could significantly reduce GLU, TC, TG,and significantly increase HDL-C in type2diabetic mice.3.The double evaporate water(Fral) of Momordica Chanrantia extract could significantly reduce AST、ALT in type2diabetic mice. 4.Pathological alterations in the livers, the livers histopathologic changes were markedly improved in the double evaporate water(Fral) of Momordica Chanrantia extract treated animals by the evidence of fewer hepatic lipid droplets and less lipid inflammatory area.5. After dosing, the double evaporate water(Fral) of Momordica Chanrantia extract can significantly reduce the relative expression of G6pc and can significantly increase the relative expression of GCK and CYP7A1,promoted the glucose metabolism.Conclusion:Significantly, the present study found that the different kinds of Momordica Chanrantia extract prevented hyperglycemia and dyslipidemia in type2diabetic mice. Clearing the double evaporate water(Fral) of Momordica Chanrantia extract is the best bioactive in hyperglycemic. And the PCR techniques was wonderfully used in hypoglycemic mechanism. Open up new ideas for the traditional Chinese medicine in the treatment of diabetes. In addition,fuether comprehensive pharmacologic investigations, including experimental long-term studies, should be carried out on its hypoglycemic mechanism.
