Objective To investigate the effect of multiple organ dysfunction syndrome (MODS) patients’serum on human umbilical vein endothelial cell function, to study the antioxidant and protective role of glutathione in human umbilical vein endothelial cell function.Methods Patients met the diagnostic criteria for MODS were included in the study. The serum stored at-80centigrade after blood samples were drawn with centrifugation within24h. The basic information of included patients were recorded, and biochemical parameters, arterial blood gas analysis, APACHE Ⅱ score, SOFA score,28days mortality, and other indicators were evaluated. Human umbilical vein endothelial cells (HUVEC) were cultured, and the cells were divided into4groups: control group, serum treatment group, N-acetylcysteine (NAC) group, GSH treated group (6mg/ml、12mg/ml、18mg/ml). Enzyme activities including dismutase (SOD), glutathione peroxidase (GSH-px), total antioxidant capacity (T-AOC), catalase (CAT), and malondialdehyde (MDA) were determined by biochemical methods. The cell viability evaluated by MTT assays. After stained by chloromethyldichlorosilane fluorescein diethyl (CM2-H2DCFDA), intracellular reactive oxygen species (ROS) strength was detected by the CM2-H2DCFDA oxidation products dichloro-fluorescein with flow cytometry. Each measured value with mean±standard deviation (x±s). The SPSS16.0statistical analysis software was used to analyze the data. The two groups’data were compared using t test, and multiple groups were compared using q test. p<0.05was considered statistically significant.Results20patients with MODS were included in the clinical study, which12cases survived, and8cases died. The age, APACHE Ⅱ score, SOFA score was significantly higher in the death group than the survival group. The survival patients’ albumin, arterial oxygen pressure (PO2) and oxygen index (PO2/FiO2) were higher than those of the death group, while the creatinine, urea nitrogen, blood lactate were significantly higher in the death group than the survival group. In the cell experiment, the level of SOD, GSH-px, T-AOC and CAT was higher after the cells were dealt by serum of patients with MODS, while these parameters were higher than those of serum group sincere GSH pre-treatment. The endothelial ROS and MDA levels were increased in the serum treatment groups, but decreased in the GSH treatment group. In addition, MTT cell viability was measured showed, and the result showed that cell activity in serum treated group was significantly lower than the control group, and increased cell activity after GSH pre-treatment.Conclusion The endothelial cell function was impaired in patients with multiple organ dysfunction syndrome. Glutathione may protect the endothelia cell by reducing free radical formation, inhibiting lipid peroxidation, enhancing cell activity.
