Aim To investigate Epstein-Barr virus (EBV) infection in gastric cancer in Wuwei area of Gansu province and the roles of miR-101, EZH2, COX-2in EBV-associated gastric carcinomas (EBVaGC).Methods Tissue microarray, immunohistochemistry and in situ hybridization were used to detect the expression of EBV-encoded small RNAs (EBERs), miR-101, EZH2and COX-2in gastric cancer tissues (n=120) and corresponding adjacent tissues of gastric cancer (n=120).Results The positive rate of EBV was10.0%in120cases of gastric cancer tissue, but EBER was absent in the corresponding adjacent tissues of gastric cancer. The difference of the EBV positivity were significant between cancer and adjacent tissues (P<0.05). EBVaGC was not significantly associated with lymph node metastasis, developed most often in cardia and body (P<0.05). The positive rates of miR-101, EZH2, and COX-2in120cases of gastric cancer tissues were8.33%,78.33%, and64.17%, the positive rates of miR-101, EZH2, and COX-2in corresponding adjacent tissues, the difference of their positive rates were significant(P<0.05). The positive rates of miR-101, EZH2, and COX-2in12cases of EBVaGC were41.67%,50.00%, and33.33%, the positive rates of miR-101, EZH2, and COX-2in108cases of EBVnGC (EBV negative gastric cancer) tissues were4.63%,81.48%, and67.59%, the difference of their positive rates were significant (P<0.05). The infection of EBV in gastric cancer tissues was related to miR-101expression positively, miR-101expression was related to lymph node metastasis, expression of COX-2and EZH2in EBVaGC tissues negatively (P<0.05).Conclusion The infection of EBV was related to part of the occurrence of gastric cancer in Wuwei area of Gansu province. EBVaGC was not significantly associated with lymph node metastasis, developed most often in cardia and body. The down-regulated of miR-101and the up-regulated of COX-2,EZH2, may be one of the reasons that contribute to the low lymph node metastasis rate of EBVaGC.
