Usingintelligent non-invasive blood pressure measurement in the rats initial blood pressure and after treatment for4hours,2weeks,4weeks,6weeks rattail artery systolic blood pressure changes, each measurement time for the experimental intervention after4hours.Continuous treatment for6weeks.After the end of treatment,the collected24hours urine of rats,with ELISAmethod to measure urine trace albumin in the urine(mALB),urine beta2-microglobulin(beta2-MG) content;Abdominal aortic blood, part of blood serum,centrifugal separation using fully automatic biochemical analyzer todetectthe contents of serum creatinine and urea nitrogen; Portions of the bloodwith anticoagulant, centrifugal plasma collection, with ELISA method todetect the plasma nitric oxide (NO) and the top ring element (PGI2), angⅡ(AngⅡ) content; In kidney tissue50mg in pathological morphology observation and HE staining immunohistochemical method to detect the expression ofcollagen type â… and â…£; Another kidney tissues around50mg with RT-PCRmethod to detect renal kallikrein mRNA, TGF beta1mRNA expression.Data is the single factor analysis of variance with the SPSS16.0software,comparing two groups using the LSD method analysis,and results expressedas mean±standard deviation (x±s),P<0.05was considered significant difference.Results:1.Acupuncture Zusanli and Quchi on SHR about systolic blood pressure has good antihypertensive effect, similar to the antihypertensive effect of perindopril treatment group, and acupuncture instant depressurization effect ofwestern medicine group.2.Acupuncture Zusanli and Quchi improved SHR plasma vascular endothelial active substances NO, PGI2levels,reduced the conten of vasoactive substancesAngII,adjusted Vascular Endothelium balance function.3.Acupuncture Zusanli and Quchi significantly lower the contention in SHRserum urea nitrogen(BUN) and creatinine(Scr) and the urinary beta2-micro-globulin(beta2-MG) and of albumin (mALB) in urine, kidneyfunction improvedsignificantly.4.Acupuncture Zusanli and Quchi improve the SHR kidney tissue pathological structure, reducing the expression of of collagen typeâ… and IV, reduce glomerulosclerosis and renal interstitial fibrosis.5.Acupuncture Zusanli and Quchi raise the level of SHR kallikrein mRNA expression, reduce the levels TGF-beta1mRNA expression, play a role of kidneyprotection.Conclusion:1.Acupuncture Zusanli and Quchi has antihypetensive effect in SHR.2.Acupuncture Zusanli and Quchi in SHR plays a protective role of hypertensiverenal impairment.3.Acupuncture Zusanli Quchi through the renin-angiotensin system (RAS) andkallikrein kinin systesm (KKS) regulate vascular endothelial diastolic factorof NOã€PGI2ã€Angâ…¡ã€kinin release equilibrium and reduce the TGF-beta1expressionof SHR for hypertension and renal protection. |
