Maozhiqing prescription, composed of Radix Ilex Pubescens、Radix Scutellariae、Herba Houttuyniae、Radix Isatidis and Folium Isatidis, with the effects of Qingrejiedu, expectorant and relieving cough, and promoting blood circulation to remove blood stasis, is mainly used to treat pediatric pneumonia. This subject developed the traditional Chinese medicine prescription into Maozhiqing oral liquid, built the quality control standards, optimized the preparation technology, and made the anti-inflammatory action study.Four compounds were isolated from Radix Ilex pubescens from Guangxi, identified as ilexgenin A, ilexsaponin A1, ilexsaponin B2, tortoside A. With tortoside A (isolated from Radix Ilex pubescens from Guangxi), baicalin, indirubin, methyl nonyl ketone as reference substances separately, the subject built the TLC identification methods and the HPLC (or GC) content determination method of Radix Ilex Pubescens, Radix Scutellariae, Radix Isatidis and Folium Isatidis, Herba Houttuyniae. The methods can be used for quality control of Maozhiqing oral liquid because of their simplicity, accuracy, high sensitivity and good reproducibility.With volatile oil content, indirubin content and dry extract rate, baicalin content、tortoside A content and dry extract rate as evaluation indexes separately, orthogonal test was used to study extraction process of volatile oil from Herba Houttuyniae, the alcohol extraction process of Folium Isatidis, and the water extraction process of Radix Ilex Pubescens, Radix Scutellariae, Radix Isatidis. With baicalin content and tortoside A content as evaluation indexes to study the alcohol precipitation process. The best preparation technology of Maozhiqing oral liquid was selected. Herba Houttuyniae, adding8times amount of water, soak for1hour, extract volatile oil for4hours by steam distillation; Folium Isatidis, adding12times70%ethanol, refluxing extraction three times, every time2hours; Radix Ilex Pubescens, Radix Scutellariae and Radix Isatidis, adding ten times amount of water, refluxing extraction three times, every time2hours; Merging the water extraction liquid, adding ethanol to the alcohol content of70%after concentrating.The anti-inflammatory models of mouse auricle swelling induced by xylene and mouse paw edema induced by formaldehyde were adopted to investigate the affect of Maozhiqing oral liquid on swelling, with swelling degree as evaluation index. According to the results, the inhibition ratios on mouse auricle swelling induced by xylene of Maozhiqing oral liquid high, middle, low dose group were29.88%,17.07%and12.80%respectively. The high dose group had significant difference compared with the model group (P<0.01). The inhibition ratios on mouse paw edema induced by formaldehyde of Maozhiqing oral liquid high, middle, low dose group were20.88%,16.37%and18.10%respectively. The three groups were extremely significant differences, compared with the model group (P<0.01).To analysis preliminary anti-inflammatory mechanism of Maozhiqing oral liquid by deteminating the PGE2levels on mouse inflammatory foot, with ultraviolet-visible spectrophotometer used. According to the results, The PGE2levels on mouse inflammatory foot of Maozhiqing oral liquid high, middle, low dose group were all less than model group. The high-dose group had significant difference compared with model group (P<0.05). The results suggested that inhibiting the generation and release of PGE2on inflammatory tissue maybe one of anti-inflammatory mechanism of Maozhiqing oral liquid.
