Swertia mussotii Franch. is Gentianaceae swertia plant and one of Tibetan medicine Zangyinchen plants.as one-year plant, distributed at an altitude of1900-3800m in Tibet, Yunnan, west northwest of Sichuan, and Southwest of Qinghai, is the raw material of medicinal used in the treatment of diseases of the liver and gallbladder system. The compounds containing in Swertia mussotii include ketone, sweroside, gentiopicroside etc.The L16(45) orthogonal experiment design and single factor analysis were utilized to select the most effective technology of Swertia mussotii compound extraction. Four levels and five factors including ethanol concentration, extraction times, material liquid ratio, extraction time, and extraction temperature were designed and studied. HPLC method was used to analyze content of the sweroside and gentiopicroside. Results showed that90%ethanol as extraction solvent, material liquid ratio1:24, boiling, reflux extracting3times, and4h were one of the most effective technology, which was stable and feasible.This experiment was studied with the polyamide and silica gel chromatography for separation and purification. According to the physical and chemical properties and spectral data, four kinds of compounds which were1-hydroxy-3,7,8trimethoxyxanthone,1-hydroxy-2,3,4,5-tetramethoxyxanthone,1,8-dihydroxy-3,7dimethoxyxanthone,1,7,8-trihydroxy-3-methoxyxanthone were separated and identified.
