Objective:to investigate the association between A1298C mutation at MTHFR gene and primary hyperuricemia of the staff working in Yangzhou University.Methods:A case-control study was conducted with237cases, which containing about116cases of hyperuricemia and121cases as matched control. All the cases were selected from the Medical Center of Yangzhou University during April-May,2010. Individual anthropometric and biochemical variables were measured. Combined by higher concentration agarose gel electrophoresis, the allele and genotype among each group were compared. The relationship of the clinical biochemical results, gene polymorphism and the pathogenesis of hyperuricemia were studied by Logistic regression analysis.Results:(1) There are not significant difference in age and LDL between Control and HUA group (P>0.05); and there are significant difference in BMI, SBP, DBP, FPG, TG, urea nitrogen, TC, HDL, UA between Control and HUA group (P<0.05),especially in BMI, SBP, DBP, FPG, TG, urea nitrogen, HDL, UA (P<0.01).(2)Logistic regression analysis suggests that BMI, SBP and TG are risk factors of hyperuricemia.(3) There are three kinds of genotypes AA, AC, CC in the two groups, the frequencies of genotype AC+CC are not significantly higher in group HUA than that in group Control(P>0.05).(4) There are A, C alleles and AA, AC, CC genotypes in the237cases. The mutational C allele frequency is15.1%in HUA and15.7%in Control group, which are not significantly different(P>0.05).(5) In group HUA and Control, the uric acid levels of AC+CC genotypes are not significantly higher than that of AA genotype(P>0.05).(6) There are not significant difference in BMI,SBP and TG between the cases with C allele and those without C allele both in HUA and Control group.Conclusions:(1) BMI, SBP and TG are risk factors of hyperuricemia.(2)There is MTHFR A1298C polymorphism in the staff working in Yangzhou University, and the genotypes are AA,AC and CC.(3) MTHFR A1298C polymorphism is not associated with the pathogenesis of hyperuricemia, the C allele is not a risk factor.(4) Maybe the MTHFR A1298C polymorphism has more influence on the regulative effects but not on the activity and function of MTHFR.
