Tu-niuxi has the effects of blood stasis, diuretic, alexipharmic. According the Traditional Chinese Medicine Standard of Hunan Province of2009edition and Traditional Chinese Medicine Pieces Processing Norms of2010, Tu-niuxi is derived from the dried root of Achyranthes aspera Linnaeus, which belongs to Amaranthes aspera. It Mainly used for gonorrhea, hematuria, amenorrhea, rheumatoid arthritis, beriberi, edema, malaria, dysentery, diphtheria, carbuncle swollen, traumatic injury and mainly distributes in Hunan province, Jiangxi province, Fujian province, Taiwan province, Guangdong province, Guangxi province, Sichuan province, Yunnan province and Guizhou province. India, Vietnam, Philippines, Malaysia also have distribution. The material basis of research on the dried root of Achyranthes aspera Linnaeus was less, the chemical constituents contained mainly for insect hormone, triterpenoid glycosides (aglycone is oleanolic acid). In order to make full use of plant resources, develop the medicinal value and further development of Achyranthes aspera Linnaeus, it is necessary to study the pharmacological activity and chemical composition. This paper contents include:Part oneStudying on the antibacterial activity and anti-inflammatory activity of different extracts of Achyranthes aspera Linnaeus screen the pharmacological active part to guide the extraction and separation of compounds.Study on the antibacterial effect of different extracts of Achyranthes aspera Linnaeus by determining the MIC of extracts with two times dilution determination using Staphylococcus aureus, Bacillus subtilis and Pseudomonas aeruginosa as target bacteria. Experiments show that: n-butanol extract and water extract of Achyranthes aspera Linnaeus had no antibacterial activity against Staphylococcus aureus, Bacillus subtilis and Pseudomonas aeruginosa. That is nothing to do Qingre and detoxifying effect with bacteriostatic of Achyranthes aspera Linnaeus. Its principle may relate to complex components of traditional Chinese medicine and human mechanism complexity, which is worthy of further study and discussion.The anti-inflammatory experiments of n-butanol extracts of Achyranthes aspera Linnaeus concluded mice ear-swollen induced by xylene and egg white-induced toe swollen. Experiments show that: n-butanol extract of Achyranthes aspera Linnaeus has obvious anti-inflammatory effect on mice auricle swelling induced by xylene. That n-butanol extract mainly contains saponins have anti-inflammatory effect. As for the anti-inflammatory mechanism, it is worth further study.Part twoIsolation and structure identification of the chemical constituents of Achyranthes aspera LinnaeusThe dried roots of Achyranthes aspera Linnaeus were extracted with70%alcohol. The extracts were concentrated in vacuum and then extracted in ethyl acetate and n-butanol, successively to give different fractions. These fractions of the dried roots were separated and purified by Silica gel column to obtain7compounds. The structures were identified by spectral evidence of NMR and MS. These compounds are identified as daucosterol (Ⅰ),oleanolic acid (Ⅱ), ecdysterone (Ⅲ),25-S-inokosterone (IV),25-R-inokosterone (V), oleanolic acid-28-O-β-D-glucopyranoside (Ⅵ) and compound7(Ⅶ). In addition to, daucosterol,25-S-inokosterone,25-R-inokosterone, oleanolic acid-28-O-β-D-glucopy-ranoside were obtained from Achyranthes aspera Linnaeus for the first time.Part three:Analyze of chemical constituents of Achyranthes aspera Linnaeus and Hou Yanqing oral liquid which contains Achyranthes aspera Linnaeus.Established a method for the determined of oleanolic acid-28-O-β-D-glucopyranoside in Achyranthes aspera Linnaeus by RP-HPLC method firstly. The method is simple accurate and reliable, which can be used for the quality control of Achyranthes aspera Linnaeus.Established a method for the determined of oleanolic acid-28-O-β-D-glucopyranoside in Hou Yanqing oral liquid by RP-HPLC method firstly, which can provide reference for quality control of Hou Yanqing.
