| Lidantang is a clinical experience decoction of our hospital surgery under the guidance of the basic theory of TCM, combined with years of experience in treating cholelith disease. Confirmed by clinical observation, it has good cholagogic and the prevention and control of cholelith disease. Lidantang is mainly composed of Chaihu, Yinchen, Zhishi, Muxiang, Jinqiancao, Baishao,Houpu, Daihuang, Wuyao, Chuangxiong and Zhigancao, etc, which has the effect of soothing-liver and regulating qi, dispersing stagnated liver qi and discharging calculus, clearing heat and eliminating dampness, etc; which has obvious curative effect to the gallbladder calculus cholecystitis (Qi-stagnation of Liver and Gallbladder and part of Damp-Heat in Liver and Gallbladder);which can effectively improve the patient’s clinical symptoms, especially the right rib and right upper quadrant pain. Therefore, this experiment, by observing variations of amount of bile secretion, total bile acid, total bilirubin and total cholesterol in bile on normal rat, studies the cholagogic effect and the mechanism of prevention and cure cholelith disease; which aims to fully explore the pharmacological effect of Lidantang and provide theoretical basis for clinical application.Objective:To investigate the cholagogic effect of Lidantang on Wistar rats and provide theoretical basis for clinical application.Methods:A total of50Wistar rats were randomly divided into five groups: saline group, Danningpian group (0.54g/kg) and Lidantang groups (6,12,24g/kg). There were ten rats in each group. Each group was intragastric administration for three days and was fasted12h before the experiment. At the start of the experiment, after anaesthetizing rats by injected10%chloral hydrate4ml/kg from intraperitoneal, fixed the rat on its back to the laboratory animal operating table, and make a incision about3cm in the midline of upper abdomen, Open into the abdominal cavity step by step, found white transparent toughness bile ducts in the descending part of duodenum; and then isolated the bile duct away from duodenal papilla about2cm, inserted the closed venous indwelling needle in diameter of0.7mm in the direction of the liver, pull out the needle core, saw needle tubing with pale yellow bile flow, bile was collected by1.0ml centrifuge tube. Staying the operation model of rats stable for20min, began to collect the bile, the bile volume were measured at1,2,3and4hour after draining the bile; the total bile acid, total cholesterol and total bilirubin were analyzed.Results:The volume of bile were significantly increased in the Lidantang groups in compared with the saline group (P<0.01), especially the high dose group making the most significant difference; Danningpian group also increased significantly (P<0.01), less effective than Lidantang high dose group;2. Compared with blank control group, Lidantang in low, middle and high dose groups each had a certain degree of increase in bile acid content in bile, while bilirubin and cholesterol levels had a certain degree of reduction, in which the difference among each group the high dose group was the most significant. It showed Lidantang can promote not only the effect of increased levels of bile acid in the bile, and reduce the content of the cholesterol and bilirubin in bile.Conclusion:Lidantang has stronger cholagogic effects on normal Wistar rats and could increase the content of the total bile acid in the bile while decreasing the content of total cholesterol and total bilirubin. |
