Objective: In this study, we established an ovariectomized rat model, to explore the effects andintervention of aerobic exercise on expression of IRS-1in skeletal muscle of ovariectomized rat.Methods: According to their body weights. The487-month-old female SD rats, were randomlydivided into four groups with each12rats: SHAM, OV, Ex0, Ex48. One week after being trainedadaptively for10min/day, the exercise groups would be trained by swimming for12weeks and it’snon-weight bearing, they did swimming exercise at the previously made program with6days per week.After the experiment, measured body weight in rats again; measured blood glucose and triglycerides byfully automatic biochemistry instrument; measured blood insulin by ELISA testing. assessed expressions ofIRS-1in skeletal muscle of ovariectomized rat by western-blotting.Results:(1) The groups were breeding and training for12weeks, their body weight increasedsignificantly, amounts of weight gain in SHAM and the exercise groups was lower than OV remarkablely(P<0.01), but there was no significant difference between the exercise groups (P>0.05);(2) Comparedwith SHAM, the triglyceride in OV was significantly higher (P <0.01), and in the exercise groups lowerthan OV remarkablely (P <0.01), there was no significant difference between the exercise groups (P>0.05);(3) The blood glucose in OV is higher than the other three groups (P <0.01), and Ex0was significantlylower than Ex48group in blood glucose (P <0.01);(4) Compared with OV, INS levels in the other threegroups were significantly lower (P <0.01);(5) The expressions of IRS-1in skeletal muscle in OV wassignificantly lower than the other three groups (P <0.01), and Ex0was slightly higher than Ex48(P <0.05). Conelusions:(1)After ovary removal, the amounts of weight gain, TG, blood glucose and blood insulinlevels were changeed and significantly higher than sham, the expressions of IRS-1in skeletal muscle wasreduced significantly, This could cause insulin-resistance easily;(2) Aerobic exercise could certain releasebody weight increased significantly, lower triglyceride, blood glucose, blood insulin levels, and couldeffectively improve the glucose-lipid metabolism disorder.(3) Aerobic exercise could effectively increasedthe activity and expressions of IRS-1in skeletal muscle,and help to improve insulin resistance. The effectsof long-term regular aerobic exercise on the activity and expressions of IRS-1in skeletal muscle had timephase property, and this effects of expressions of IRS-1was more obvious immediately after long-termregular exercise.
