| Objective: Sports fatigue mechanism has been always highlighted in the field of sportsscience research. With the deepening research of sports fatigue, scientists shifted theirresearch orientation gradually from traditional peripheral fatigue to central mechanismof sports fatigue. GAP-43, specific protein phosphate being widely indwelled uponnerve cell membranes, sharing significant relevance with neural development, axonalregeneration and synaptic reorganization, nowadays has been preferred the exclusivemolecular probe to study neural plasticity of nerve sprouting, neural development anddamage healing,etc. the changes in the structure and function of the nervous system,inthe neurotransmitter and neuromodulator caused by sporting pivot fatigue could havedirect or indirect effects to the GAP-43expression, however, there are few studiesregarding the GAP-43different expression and learning and memorizing changes in thecircumstances of extreme loading sports(ELS),. This article could provide a theoreticalbasis for the general public appropriate physical exercises and rehabilitations bystudying the GAP-43genetic and protein expression changes in sporting pivot fatiguesand the effectiveness of extreme sports to learning and memorizing abilities in thebrains.Methods: Behavior training for ten days have been taken to the SD rats in Morrisconstant temperature water maze, and then let the adult rats with5%of its own weight swim for one time ELS; observe dynamic changes by the ELS to adult rats learning andmemorizing effectiveness. The adult rats are randomly divided into different grouping,namely blank group(C), water maze training(WMT)&ELS scheduled drawn group(O),WMT&ELS drawn group and WMT group. The WMT&ELS scheduled drawn groupdivided into0-hour group(OW0),12-hour group(OW12)24-hour group(OW24),48-hourgroup(OW48), after10days of WMT, then ELS and behavior training. The WMT groupdivided into0-hour (W0) group,12-hour(W12) group,24-hour(W24) group,48-hour(W48)group. HE staining, real-time PCR and western-blotting and other methods have beenapplied to study adult rat and hippocampus GAP-43dynamic changes.Results:1.Water Maze Latency Training: after ten days of water maze training, thelatency period of the adult rats are significantly shorten and latency period of the tenthday apparently different from the first day of each group, P <0.05, which states thewater maze training could have the adult rats formed spatial memory; The test resultsbetween10thday and after10thday of the WMT group, the W0,W12,W24,W48group a bitslightly increase, however, no big difference between groups,>0.05, the run-acrosstimes counted not much changes.2. HE Staining Observation: the hippocampus tissue ofthe matched group structural integrates while the ELS ones structure looses withdecreased density and cell vacuoles, which implies that the ELS could cause someextent damages to hippocampus tissue and structures.3. GAP-43Protein TestingResults: comparing to the matched group, the OW0ã€OW12ã€OW24group after once ELShave very different GAP-43protein expression quantity descending outcomes, OW48stays almost the same; but comparing to the other ELS treated groups, OW48has verybig changes,P<0.01; comparing to the matched group the OW0ã€OW12ã€OW24groupafter once ELS have very different GAP-43protein expression quantity descendingoutcomes, OW48stays almost the same; but comparing to the other ELS treated groups,OW48group obviously increased a lot and has very big changes, P<0.01; the WMTadult rats’ hippocampus comparing to the matched group, the GAP-43goesdownwards first then upward, while the ELS groups have stronger performance.Conclusion:1. ELS one time could effect the hippocampus GAP-43genetic proteinexpression after adult rats’ WMT, and the GAP-43expression trended first going downlater going up; the WMT group shared almost the same, going down first then up, withnot-so-strong intensity than the ELS, which explicates the GAP-43possibly couldrenovate and regenerate the damaged nerve cell and such expression seldomlyperformed in a more matured nerve cell.2. ELS once could cause weakening spatialcognition, extraction of spatial memory, and such effectiveness presented very obviousafter ELS, but could recover itself after12hours. |
