Abatract: Objective: The latest Nobel Prize research found that the brain has a GPS(navigation systems), American scientist John ? O’Keefe found to let people know where they are in a "place cells", further research from O’Keeffe also found that all "place cells" are concentrated in the brain of mice, "hippocampus" in a particular area. and "place cells" in the "Hippocampus" is a member of the brain GPS for spatial cognition and positioning capabilities playing an important role, but there is little relevant research to study whether the spatial localization ability is relevant to the nerve cells energy metabolism, this paper on spatial orientation and learning about the work GLUT4 investigate whether a memory ability and hippocampus involved in energy metabolism, and viewed whether through moderate-intensity exercise training can improve spatial orientation and SD rats learning and memory capacity, improve SD rat hippocampus SD rats glut4 expression.Methods: testing 32 8-week-old SD rats’ s learning and space memory ability by water maze experiment, divided into Good and Poor group in accordance with the spatial orientation and learning and memory ability. the best record for a group of eight Morris water maze good group(MG group), the worst record for a group of eight Morris water maze poor group(MB group), Western blotting detect protein expression of hippocampus glut4 for MG group and MB groups;the remaining 16 are divided into two groups(follow two group of spatial orientation and learning and memory scores mean difference between the minimum, P values of independent samples T-test of the principle of the better groups), were recorded as the Morris water maze training group(ME) and Morris water maze in the control group(MC), ME group with moderate intensity weight training swim for five weeks, MC group consisting of eating five weeks, positioning and after five weeks test spatial orientation and learning and memory ability with the Morris water maze, detect protein levels in the hippocampus glut4 by Western blotting.Results:(1)Average incubation period of the Morris water maze test(19.42±1.72)from strong group with untrained andgood innate spatial orientation and learning ability than the that group with untrained and bad innate spatial orientation and learning ability(42.67±4.61) had significant difference(P = 0 < 0.01);(2) GLUT4 expression levels of hippocampal cell from strong group with untrained and good innate spatial orientation and learning ability(4.04±1.4) was significantly higher than the that group with untrained and bad innate spatial orientation and learning ability( 1.67±0.49), P=0.029 < 0.05;(3) Comparison of spatial orientationand learning ability between ME group with 5 weeks of moderate-intensity swimming training and MC group with untreated swimming training, there is the existence of significantdifferences, on escape latency time everyday, ME group(dl: 14.25± 6.85; d2: 14.64±7.78; d3:17.83±10.44;d4 10.98±5.94)is shorter than the MC group(dl:15.48±4.69;d2:15.55±7.83;d3:20.26 ±11.67;d4:14.89±6.29), as to the number ofacross platforms, ME group(3.87± 1.36)is more than MC group(2.718±0.99), the percentage of distence traveled of objective area from total distance in two groups were compare had significant difference by the Pearson Chi-square Test(X2=15.864,P=0), Space exploration path chart also show: swimming trajectory of control MC rats ware scattered or stick wall, the experimental group ME rats are more concentrated in the target quadrant, In a word, the experiment results of the navigation and space exploration,shows that the experimental group ME and control group MC had significant difference in the spatial orientation and learning ability.Conclusion: The Space(1) position of learning and memory in rats with hippocampal GLUT4 involved in energy metabolism related;(2) exercise can improve spatial orientation and learning and memory in rats,Speculated that this may be related to movement increase expression of GLUT4 involved in energy metabolism inrats.
