Objective: Application of aerobic exercise (60min/d swimmingtraining) in rats induced by d-Galactose in senile model, determination of aerobicexercise on SD rats with brain relating indicators of aging and its effects on learningand memory ability and hippocampal GAP-43gene and protein expression data, andlocation. Study on aerobic exercise on brain aging and learning and memory abilitiesof rats and the effect of GAP-43gene and protein expression in hippocampus.Methods:84SD rats were randomly divided into3groups: acute senile Group (D),and the saline control group (Na group), acute aerobic exercise as a part of agingintervention group (Ds). ND set of natural feeding1month before injection ofd-Galactose6weeks,Na set of natural feeding1month before injecting0.9%Nacl,6weeks, Ds group of natural feeding1month after injection of d-galactose and60min/dswimming training for6weeks. Each group were randomly divided into2groups: agroup doing7days Morris water maze test marked DsM and DM, NaM groups group,another group of natural feeding for7days, marked as DN, NaN groups, DsN group,divided into6groups. Determination of extraction of rat brain tissue in rats of eachgroup activities of brain SOD, GSH-PX and MDA content, test whether aging modelinduced by d-Galactose model successful and aerobic exercise on the activity of SODand GSH-PX and MDA content of. Morris water maze test on learning and memory inrats of each group, Real-time PCR, Western-blot, immunofluorescence detection inhippocampus of rats of each group of GAP-43gene and protein expression andlocalization. Results:1. Morphological observation of results: D group compared withthe Na group, looking sleepy, sluggish, lethargy, curly fur Brown, without gloss, agingsymptoms such as severe shedding of hair.2、Radicals test results: SOD activity issignificantly lower than the DM group NaM Group (P<0.01), significantly belowNaM GSH-PX activity group (P<0.05) and the MDA content is significantly higherthan the NaM Group (P<0.01). DsM SOD, GSH-PX activity of the Group wassignificantly higher than the DM Group (P<0.01) and the MDA content wassignificantly lower than the DM Group (P<0.01). DsM and NaM group activity ofSOD and GSH-PX and MDA content were not significantly different (P>0.05). SOD,GSH-PX activity significantly below NaN DN Group (P<0.01), content of MDAsignificantly above NaN group (P<0.01), DsN SOD activity is significantly higherthan the group DN group (P<0.01) and gsh-px activity than DN group (P<0.05) andthe MDA content is significantly below the DN group (P<0.01), DsN and NaN groupactivity of SOD and gsh-px and MDA content were not significantly different (P>0.05).3、Morris water maze navigation test results: NaM and DsM group2nd dayprimary formation of spatial memory, DM Group3rd day of spatial memoryformation, NaM3rd day to form a stable set of spatial memory, DsM4th day to forma stable set of spatial memory. The space exploration Results: NaM1th DM groupquadrant platform into the water through the very significant decrease in the numberof (P<0.01); DM1th DsM group quadrant platform into the water through asignificant increase in the number (P<0.05).4, Hippocampus GAP-43mRNA testresults: NaM DM group group of GAP-43mRNA expression in hippocampus of avery significant drop (P<0.01); DM DsM group group expression very significantincrease (P<0.01); NaM DsM group expression significantly raised in comparison(P<0.05). NaN DN group GAP-43mRNA expression in hippocampus of a verysignificant drop (P<0.01), DsN group DN group expression are significant increases(P<0.01); NaN DsN group notable decline in group expressions (P<0.05). DM DNgroup set, DsN DsM group set of mRNA expression were not significantly different(P>0.05).5, Hippocampus GAP-43protein test results: DM NaM group GAP-43protein expression in hippocampus of a very significant drop (P<0.01); DM DsMgroup group expression very significant increase (P<0.01); NaM DsM group group nosignificant difference (P>0.05). NaN group DN group expression of a very significantdrop (P<0.01), DsN group DN group expression very significant increase (P<0.01),DsN NaN group expression of a significant drop (P<0.05). DM DN group set, DsNDsM group group NaN NaM group group expressed no significant difference(P>0.05).6, Hippocampus GAP-43immunofluorescence test results: GAP-43proteinpositive cells mainly in the hippocampal CA3spinal cells, GAP-43distribution on thecell membrane and bumps. DM-immunoreactive cells in the hippocampal CA3areanumber of groups less NaM group and the DsM-positive cells in the hippocampalCA3area more. DN group-positive cells in the hippocampal CA3area fewer arrangedcells scattered; NaN and DsN group-positive cells in the hippocampal CA3area largenumber arrangement scattered. Conclusions:(1) injection induced by d-Galactose6-week (100mg/kg/d)-induced aging rat model and showed obvious symptoms ofaging, aging group of brain SOD, GSH-PX activities compared to the saline controlgroup significantly reduced, significant increase in MDA content.(2) aerobic exercisewith d-Galactose induced aging processes, you can reduce the degree of impairmentof learning and memory in rats during senescence, and maintain normalGAP-43mRNA and protein expression in hippocampus. This may be slow on learning and memory impairment in rats during senescence mechanism.
