Purpose:Animal experiments using the method, by comparison with the analysisof the expression of liver tissue ChemerinmRNA/CMKLR1-mRNA movement fat dietrats under different states with a quiet level and TNF-α、MCP-1levels change,explore the effect of high fat diet and exercise anti-inflammatory effects on liverfunction in rats and the protective effect of dietary fat rat liver.Methods:Healthy8-week-old male SD rats Clean40, one week after theadaptive feeding randomly divided into normal diet does not even exercise group (N=10, C), high-fat diet without exercise group (N=10, H), normal diet exercise group(N=10, CE), high-fat diet exercise group (N=10, HE), C/CE group with normaldiet routine feeding, H/HE group of high-fat diet feeding.Weekly measurements ofbody weight, height. Motion adaptive rats for1week after treadmill exercise, aerobicexercise performed4-6weeks, using0slope treadmill speed20m/min be increased1m/min week,1/d,40-50min/times,6days/week exercise time each day:21:00-24:00.The first6weeks, the groups were fasted for12hours, the rat tail vein blood fastingglucose, blood plasma was separated after the apex30min and the supernatant wascentrifuged at3000rpm placed in vials stored at-80℃tested quickly removal of theliver, were placed in liquid nitrogen and stored in formalin for later use. ELISA methodto measure plasma cytokines TNF-α, insulin (Fins); using PCR technology to detectCMKLR1-mRNA; Western blot technique to detect Chemerin;HE staining of livertissue infiltration observed KCs. Test using SPSS11.5software processing the dataobtained.Result:1. Body weight of rats in each group are a growing trend in the course ofthe experiment. The rats in group H and group C, weight HE and CE groupincreasedsignificantly (P <0.01, P <0.05);2.Through single factor analysis of variance, high fat diet can significantly increase the plasma TNF-α、MCP-1levels in rats (P <0.01); aerobic exercise canobviously decrease the plasma levels of TNF-α、MCP-1in rats fed a high fat diet (P <0.05); aerobic exercise and high fat diet no interaction (P>0.05);3. Rat liver tissue HE staining, H group compared with C group appeared diffuse,vesicle like fatty change, visible Kupffer cell infiltration; in group HEcompared withgroup H vesicle like steatosis degree reduction;High fat diet can make the liver CMKLRmRNA/Chemerin level of rats increasedsignificantly(P <0.01),aerobic exercise can reduce the level of CMKLRmRNA/Chemerin in liver of rats fed a high fat diet(P <0.05),aerobic exercise and high fat dietno interaction(P>0.05);Conclusions:1.6weeks of high fat diet the weight of rats increased significantly,plasma levels of TNF-α, MCP-1, Homa-IR increased, leading to obesity in rats in vivo,inflammation and IR.2.6weeks of high fat diet rats, fatty degeneration of hepatocytes, infiltration ofKCs, Chemerin/CMKLR1-mRNA expression level increased, induced inflammation inrat liver.3.6weeks aerobic exercise with weight control, on the high fat diet rat reduceweight, improve insulin resistance good effect, and can reduce theexpression level ofChemerin/CMKLR1-mRNA in the liver of hepatic inflammation, alleviate to someextent, the reversal of hepatic steatosis,which has the function of protecting liver.
