| Objective:By comparing the change of IL-6, TNF-alpha level in the rats plasmaã€myocardialICAM-1ã€NF-κB-mRNAã€Apelin-mRNA before and after experiment,study the effectsof a high-fat diet on rat myocardial and aerobic exercise and its joint supplement of soyisoflavones on a high-fat diet rats myocardial protection.Methods:The clean healthy rats are48,8weeks old.Weight is180~220grams.After1weekadaptive feed, randomly divided into5groups, ordinary diet+don’t exercise group (N=10, group C); A high-fat diet+don’t exercise group (N=10,group H); A high-fat diet+aerobic exercise group (N=10, group HE); A high-fat diet+soybean isoflavonesgroup (N=6, group HS); A high-fat diet+aerobic exercise combined added soybeanisoflavones group (N=6, group HES). The HEã€HES groups are adopted zero gradientrunning machine when exercise.At the first week,the speed is15m/min.At the secondweek,20m/min. From the third week,every week the speed would add1m/min.50min/time. Time: at21:00-24:00,1time/day, six days/week, a total of6weeks. Half anhour before the exercise,according to250mg/kg, HS〠HES group are added soyisoflavones.Other groups will be added equal volume of saline. Weekly,test weightand length data,then record. After the experiment, use the method of Elisa testing ratsplasma IL-6, TNF-alpha level and the levels of myocardial ICAM-1, PCR testmyocardial NF-kappa BmRNA, Apelin-mRNA levels. Using SPSS11.7softwareprocessing obtained data.Results:1. Lee group H rats’ s index, serum TNF-É‘, IL-6levels, myocardial Apelin- mRNA,NF-кB-mRNA expression level and ICAM-1content is significantly higherthan group C (P <0.01, P <0.05).2. Aerobic exercise, soybean isoflavone supplements may reduce the high fat dietrats lee ’s index (P <0.01, P <0.05); There was a significant interaction between the two(P <0.01), meaning that the same movement condition, lee added soy isoflavones canmake the rats’ s index significantly decreased (P<0.05).3.Aerobic exercise can significantly reduce the high fat diet rats plasma TNF-É‘,IL-6levels (P <0.05, P <0.01); Add soy isoflavones can decrease the levels of serumIL-6(P <0.05), the level of serum TNF-É‘ effect is not obvious (P>0.05); Nointeraction between them (P>0.05).4. Aerobic exercise, added soybean isoflavones to reduce the high fat diet ratsmyocardial Apelin-mRNA, NF-κB-mRNA expression level and ICAM-1content hasa significant effect (P <0.01); To reduce ICAM-1content between significantinteraction (P <0.05), that is, at the same time supply under the condition of soybeanisoflavones, aerobic exercise can make rats ICAM-1significantly decreased (P<0.05);To reduce the myocardial Apelin-mRNA, NF-κB-mRNA expression level withoutinteraction (P>0.05).Conclusions:1.6weeks high fat diet can increase the weight of rats and lee ’s index in differentdegrees, serum TNF-É‘, IL-6levels, trigger inflammation in the body.2.6weeks in rats caused by high fat diet can increase myocardial Apelin-mRNAand NF-κB-mRNA expression, ICAM-1content increases, the myocardial inflammationreaction, myocardial vascular endothelial damage.3. Aerobic exercise or soy isoflavones can reduce weight, reduce inflammation andalleviate myocardial inflammatory reaction, thus protecting myocardium; Aerobicexercise combined added soy isoflavones on myocardial protection than simple aerobicsor added soybean isoflavones more effective. |
