Objective:Choose male SD rats as the experimental subject,giving swimmingtraining after spinal cord injury in rats.At different time to assess the movement functionof spinal cord injury rats,measure the content of serum,observe tissue morphologicalchanges and make quantitative analysis for the expression of basic fibro blast growthfactor(bFGF) protein in the injuried rats through immunohistochemicaltechnique.Through the results of statistical analysis of these indicators,to illustrate theeffect and possible mechanism of swimming training on spinal cord injury recovery.Methods:Used modified Allen’s strike method manufacturing rat spinal cord injurymodel.90male rats were randomly divided into sham group(n=30),natural healinggroup(n=30)and swimming training group(n=30).Then they were divided into afterinjury1dgroup(n=6),7d group(n=6),14d group(n=6),21d group(n=6),28dgroup(n=6)depending on different sampling time.The rats in swimming training groupbegin to have swimming training in the7th day after spinal cord injury. Test methodsand indicators:①Do BBB score and inclined plane test to the rats dto evaluate the motorfunction in rats with spinal cord injury.②Take blood in rats’heart at different time andby colorimetry to measure the content of calcium,potassium and the enzyme activity ofAST and ALT in the rats’serum. Then analyze the statistics after SCI.③Observe themorphological changes in the rats’spinal cord tissue after HEdyeing.④Immunohistochemistry technique to detect the antigens distribution of bFGFin the injured spinal cord tissue and observe the protein expression of bFGF.Results:①There has a sheet of bleeding in the spinal cord gray matter,cell swellingand degeneration,obvious inflammatory reaction.Swimming training group than in thegroup of natural healing after modeled(14d、21d、28d)improved significantly,the spinal cord tissue reduce bleeding, inflammatory cells decreased,intracellular basophilicparticle deposition,glial cells and a small amount of nerve fibers hyperplasia.②bFGFprotein express in injured spinal cord that:after modeled7d the expression of rats bFGFprotein increased,became reducing at28day.The expression of bFGF protein of theswimming training group significantly rises compared with the natural healinggroup(P<0.05)at after modeled14d、21d、28d.③Natural healing group and swimmingtraining rats after modeled1d serum calcium levels were significantly increased,andserum magnesium reduced.Compared with the sham group,there were a significantdifference(P<0.01).Swimming training group compared with natural healing group,in14d、21d、28d they were significantly difference(P<0.05).④After modeling1d naturalhealing group and swimming training group of AST、 ALT activity is rapidlyincreased.natural healing group were significantly increased(P<0.01)in7d,14d,21d,28dto the relative sham group.Swimming training group in21d,28d relatively naturalhealing group AST、 ALT activity significantly reduced(P<0.05).⑤The swimmingtraining group and the natural healing group after the spinal cord injury,the ramp testscores and BBB score was obviously reduced.After swimming training group modeledin14d、21d、28d,the ramp test scores and BBB scores was significantly higher than thenatural healing group(P<0.05).Conclusion:①Swimming training can improve morphological changes of injuredrats’ tissue,indicating that it promoted the recovery of spinal cord tissue in rats.②Swimming training can increase the bFGF protein expression of injuried spinalcord,boosting rats’ recovery after spinal cord injury.③Swimming training reduce serumcalcium content and increase serum magnesium content significantly,indicating that itpromotes recovery in rats.④The activities of serum AST, ALT activities in swimmingtraining group significantly reduced,suggests that swimming training has an positiveinfluence on the recovery of spinal cord injury in rats.⑤Behavioral and BBB scoreshave been enhanced through swimming training,demonstrating that it further therats’motor function.
