In this paper, the dry and mature fruit of Perilla frutescens(L.) Britt belonging to the Labiatae family was taken as the object of the research. The fractions with antioxidant activity were screened by DPPH radicals scavenging method, and the fingerprint was constructed by high performance liquid chromatography. The fingerprint-efficacy relationship between fingerprint and antioxidant activity was built by mathematical statistics method, with the combination of TLC-bioautography assay and column chromatography to determine the key components about antioxidant activity. It can provide a new idea for the quality evaluation of the fruit of Perilla frutescens(L.) Britt.Firstly, the reflux extraction was used to obtain the extracts of 22 batches of the fruit of Perilla frutescens(L.) Britt from various sources in the study. DPPH radical scavenging method was used to filter out the antioxidant activity fractions and then the antioxidant capacities of the fractions were evaluated by DPPH and FRAP assay. The chemcial fingerprint was established by HPLC to find out the common peaks. Then, the correlations between the total phenolic content, the total flavonoid content with antioxidant capacity were analysized by pearson correlation analysis. The fingerprint-efficacy relationship with the antioxidant activity fractions was established by multiple correlation analysis based on the active index of medicinal efficacy of EC50 values and the common peak areas. With the combination of TLC-bioautography assay, the four active components of the fruit of Perilla frutescens(L.) Britt were determined. Finally, on basis of the active peaks, the quality of 22 batches of the fruit of Perilla frutescens(L.) Britt from various sources were evaluated by similarity analysis and hierarchical clustering analysis(HCA). The research result shows that the key antioxidant active components of the fruit of Perilla frutescens(L.) Britt can be found through the method. At the same time, it can also provide reference for the quality evaluation of other traditional Chinese medicines.
