cAMP is a kind of cyclic nucleotide, and it is called the “second messager” in transferring life information. In China, Ziziphus jujube is famous for its various nutritions and bioactive materials, especially the higher content of cAMP, which is considered to have value of development and utilization. Taking the Ziziphus jujube as raw material to prepare the cAMP will develop new frontier in utilizing the jujube resource. This research is based on the domestic current situation of jujube resource utilization and aims to obtain the c AMP with the purity of 30% and 95%. The extraction technology, purification technology and modern analysis and detection technology were applied in this research to study the extrac tion, isolation and purification of c AMP from Ziziphus jujube. The main results were as follows:1. In this study, the cAMP from Ziziphus jujube was extracted by ultrasound-assisted method. Based on the results of single factors experiment, the orthogonal experiment was conducted and the optimum extraction conditions were obtained as follows: extraction time 12 min at normal temperature, solid- liquid ratio 1:12, ultrasonic power 240 W. Under these conditions, the yield of cAMP was 76.46%.2. The microwave-ultrasound assisted extraction was applied in the extraction of cAMP from Ziziphus jujube, and through single factors experiment, PB design and BB design, the optimum extraction conditions were obtained as follows: microwave power 440 W, microwave time 64 s, solid- liquid ratio of 1:8, ultrasonic time 21 min, ultrasonic power 300 W, and ultrasonic temperature 20 ℃. Compared with conventional water extraction and ultrasound-assisted extraction, the extracting ratio of cAMP by microwave-ultrasonic assisted method increased by 10.39% and 24.68%, respectively. Under these conditions, the yield of cAMP was 90.83%.3. D201 ion exchange resin was selected to purify the c AMP from extraction solution. The conditions of purification were as follows: concentration of solution 30 mg/L, flow speed of solution 2 ml/min, formic acid(0.5 mol/L) as desorption solution, flow speed of desorption solution 3 ml/min. We finally got the c AMP with purity of 34%.4. Studying the effect of different factors on the purification of cAMP from Ziziphus jujube by silica gel column chromatography, the conditions were determined as follows: concentration of solution 0.4 mg/mL, flow speed of solution 2 ml/min, the p H value of sample solution 4.5, ethyl acetate, benzene and methanol(1:1:3) as desorption, flow speed of desorption solution 3 ml/min. We finally got the c AMP with purity of 96%.
