Objective : In this experiment,Xinjiang jujube was used as the experimental material to establish a rapid,accurate,sensitive and reproducible HPLC assay method.The extraction process of cyclic adenosine monophosphate was optimized and the cyclic adenosine monophosphate granules and The oral liquid molding process was studied.Methods: The established method for the determination of cAMP was validated by sample recovery,reproducibility,etc.The cAMP extraction process was studied,and the effect surface experimental design method was used to optimize the extraction process,and two types of cAMP were used.The single-factor and orthogonal test of the molding process are carried out,and the optimum molding process is obtained by comprehensive evaluation and analysis of variance.Results:1.Chromatographic conditions of cAMP cyclic adenosine(cAMP)high performance liquid chromatography(HPLC): Column: Ailent HC-C18(4.6×250 mm,5 ?m)column;mobile phase: methanol-0.2% formic acid(volume Ratio 80:20);optimum flow rate: 0.8mL/min;optimal detection wavelength: 259 nm;column oven temperature 30 °C;sample volume 20 ?L.The experimental results show that the cAMP standard curve has good linearity,and the retention time of cAMP standard product is 4.7 min.The method is simple,sensitive and reproducible,and can rapidly analyze and determine cAMP content in jujube.2.Establish a method based on microwave-assisted extraction of cyclic adenosine monophosphate(cAMP)in jujube.Five factors,the ratio of material to liquid,extraction time,ethanol volume fraction,microwave frequency and microwave treatment time,were used as extraction parameters.With cAMP extraction rate as the response value,the effect surface analysis method of the central composite design was used to optimize the extraction parameters.The results are analyzed and verified.The results showed that the optimum extraction process of cAMP was as follows: material-to-liquid ratio 1:15 g/mL,extraction time 7 h,microwave frequency 270 W,ethanol volume fraction 30%,microwave treatment time 10 min,cAMP extraction rate 29.69%.3.The jujube cAMP content is high,but its product is single,the main dosage form in clinical use is cyclic adenosine monophosphate injection,its poor medication compliance,and inconvenient use and many other shortcomings,which makes cAMP Product diversification is of great necessity.A single factor experiment was conducted to comprehensively evaluate lactose,dextrin,soluble starch and the mixture of the three,and a pharmaceutical auxiliary material suitable for the preparation of cAMP granules was selected,and then the ratio of excipients and extracts and diluents were designed through orthogonal experiments.Process parameters such as ethanol volume fraction and drying temperature were optimized.The results showed that the best excipient of cAMP granules was lactose,the best molding process was 1:4 for extracts and accessories,theconcentration of ethanol was 75%,the drying temperature was 50°C,and the molding rate was over 96%.4.The single-factor test,orthogonal test and comprehensive evaluation were used to optimize the preparation process of cAMP oral liquid.The comprehensive evaluation and single factor experiments were used to screen the excipients needed in the oral solution.The orthogonal trials were used to optimize the excipients and the main ratio to optimize the oral liquid molding process.The results showed that the best process was 0.075% potassium sorbate,0.25 g maltodextrin,and the temperature was 55 °C.The cAMP content in the oral solution was greater than 3 mg/10 mL,which met the requirements of the pharmacopoeia for drug use.Conclusion: The microwave-assisted extraction method is used to extract cAMP from red jujube,its extraction efficiency is higher than other extraction methods,and the experimental operation is simple,and the extracted cAMP can be accurately quantified by HPLC method.In the cAMP molding process,The two cAMP preparations produced by the cross-test and single-factor tests have good stability,and the main content reaches the pharmacopoeia cAMP content and can be used.Therefore,the research on the molding process of cAMP in jujube has great significance for the diversification of jujube products and the economic benefits of jujube industry.
