The extraction of cAMP from Hami jujube with the different pretreat methods were studied. The effects on the different drying methods to maturity were analyzed, using BBD central composite design and response surface analysis to optimize extraction parameters in microwave and water bath, using macroporous adsorption resin for the purification of cAMP. The extraction was influenced by extraction and purification. The experimental results were as follows:(1)The gramularity of the extract had little impacts on the extraction of cAMP in fresh Hami jujube. The extraction of cAMP from syrup of dates by using cellulose was 318.21μg/g, while using pectinase was 58.42μg/g. It was proved that pectin material may protect cAMP. The content of cAMP in different maturity had a big difference, such as the green had Oμg/g, half red had 84.00μg/g, fully mature had 314.80μg/g. The content of cAMP in different parts also had difference, for example, it had 320.37μg/g in skin,251.25μg/g in meat and 23.95μg/g in nut. Drying was a predominant influence in the content of cAMP. Drying at 40℃, the content of cAMP was the most, as to 298.49μg/g, while, drying in the shade, its content was 161.94μg/g. The study also proved that brown date meat can make cAMP loss in the drying.(2)Using water bath method to extract cAMP from Hami Jujube, the optimum extraction conditions were as follows:the ratio of material-liquid was 1:44.07, extraction temperature was 89.91℃, extraction time 2h. Under such conditions, the predictive value of the cAMP extraction yield was 287.90μg/g, the measured value was 271.39μg/g, and the relative error was 5.74%.(3)Using Microwave assistance to extract cAMP from Hami Jujube, the optimum extraction conditions were as follows:the ratio of material-liquid was 1:30, extraction temperature was 81.53℃, the microwave time 7.7min, the microwave power 231.1W. Under such conditions, the predictive value of the cAMP extraction yield was 244.20μg/g, the measured value was 228.24μg/g, and the relative error was 6.53%.(4)Through to nine types of macroporous adsorption resin for static adsorption test and static desorption test, studies suggested that D101 resin and Hz802 resin were good for the separation of cAMP. The D101 resins of absorbing cAMP dynamically were as follows:Concentraction 9μg/mL, its pH5, flow rate 1mL/min. Those of desorbing cAMP dynamically were as follows:taken 25% ethanol as eluent, its flow rate 1mL/min. The Hz802 resins of absorbing cAMP dynamically were as follows:Concentraction 8μg/mL, its pH8, and flow rate 1mL/min. Those of desorbing cAMP dynamically were as follows:taken 35% ethanol as eluent, its flow rate 2mL/min.(5)The purity of cAMP was 0.10%, analyzed by HPLC (high performance liquid chromatography) method.
