Objectives Corn protein powder with poor water solubility,rough taste,utilization rate is low,limiting its application in food industry.If the corn protein powder to make a comprehensive utilization,change the above shortcomings,to apply to the development of new functional food,which effectively utilizes the main by-products produced by corn starch wet system,but also to prevent the waste of resources and environmental pollution.So the extraction subject to the corn protein powder as the raw material for the isolation and purification of lutein,and product development,and provide a theoretical basis and basic application powerful comprehensive utilization in order to corn protein powder.Methods Corn protein powder as raw material,the ultrasonic assisted extraction of lutein.On the basis of single factor test,ultrasonic power,ultrasonic time were selected as independent variables and the ratio of material to liquid,with lutein yield as response value,the interaction of each variable and its effect on the yield of lutein response surface method,get the optimum extraction parameters of ultrasonic assisted extraction of lutein;In order to obtain high purity of lutein extraction process,based on the research on saponification experiments on Extraction of lutein and lutein by C18 column chromatography purification;The application of high purity lutein microcapsule preparation,through the single factor experiment,the ratio of core to wall,the wall material ratio,inlet and outlet air temperature and dosage of emulsifier and other factors,the comprehensive effects on the microencapsulation efficiency and yield,the optimal technological conditions by the orthogonal experiment method,the optimal process parameters of natural lutein microcapsule by spray drying;The stability of colorimetric method for natural lutein microcapsules were investigated,and taking Vc as the reference,to evaluate the antioxidant activity of lutein microcapsule solution and product.Results 1、The optimum conditions of ultrasonic assisted extraction of maize protein powder of lutein:ultrasonic power 295 W,ultrasonic time 31 min,material to liquid ratio of 27:1(m L:g),under this condition,lutein was 139.65(μg/g); 2、The saponification conditions:at 25℃,the concentration of KOH-10mg/100 m L ethanol solution as saponifier,saponified 9h;column chromatography conditions: using ethanol as eluent,the flow rate was 1m L/min,the dosage of 4BV,the amount of sample solution:silica ratio of 1:100(m L:g),the final purity more than 80% of lutein; 3、In the wall material mass ratio is 15:1,the mass ratio of core to wall 1:25,feed rate,inlet and outlet air temperature of 350 m L/h were 150, 70 degrees under the conditions of preparation of lutein microcapsules,mean value of the best efficiency and yield were 89.23% and 90.16%.Lutein microcapsule product was orange powder no smell; 4、Natural lutein microcapsules were placed in high temperature,high humidity and illumination conditions,no obvious changes in the indexes of products; samples for 3 months at room temperature after the test,the results showed that the main physical and chemical indicators did not change significantly;the embedding lutein microcapsule product obtained after did not change the superoxide anion free radical,DPPH free radical and hydroxyl free radical scavenging properties,but the scavenging capacity decreased.Conclusions In this paper,the method of ultrasonic assisted extraction of xanthophylls from corn protein powder,through the single factor experiment and response surface experiments to determine the optimal extraction parameters were as follows:ultrasonic power 295 W,ultrasonic time 31 min,material to liquid ratio of 27:1(m L:g),under this condition,lutein was 139.65(μg/g);for further to obtain high purity of lutein extraction process,based on the research on the extraction of lutein,through study on separation and purification of lutein,the final purity higher than 80%;the preparation of lutein microcapsule by spray drying method,on the basis of single factor experiment,using the orthogonal experiment to optimize the process parameters,the optimal process parameters for:wall material ratio is 15:1,the quality of the core wall of the mass ratio of 1:25 and 350 m L/h,feed rate, inlet and outlet air temperature was 150,70℃,under this condition,the efficiency and the yield of lutein microcapsules were respectively 89.23%and 90.16%. Lutein microcapsule product was orange powder odorless;was studied by high temperature,high humidity index and light conditions on the natural lutein microcapsule,no obvious change;the embedding lutein microcapsule product obtained after did not change their on superoxide anion free radical,DPPH free radical and hydroxyl free radical scavenging properties but the scavenging capacity, decreased.
