In the middle of 1990 s, the concept of “bidirectional fermentation” was proposed as a production process of culture of fungus in the presence of medicinal materials, which aims to raise its pharmocodynamic effect of both resources. Panax notoginseng belongs to the family of Araliaceae Panax with traits of warm effect, and sweet and bitter taste, and has long been used as a kind of precious medicinal herb in Traditional C hinese Medicine. The active ingredients of Panax notoginseng have been determined as a class of saponins, which were confirmed to be efficient in some diseases treatment. Cordyceps sinensis is an important rare medicinal fungus. It contains many active components such as Cordyceps polysaccharide, cordycepin etc., and functions in these aspects of enhancing immunity, antitumor, antioxidant and hypoglycemic, and consequently has a great market demand.With Panax notoginseng as the medicinal matrix, and Cordyceps sinensis as medicinal fungi, the medicinal matrix with active ingredients are bio-transformed and utilized by medicinal fungi through bi-directional fermentation, thus better pharmacodynamic was achieved; in addition, the medicinal matrix could provide the necessary nutrition for the medicinal fungi. In this study, bi-directional fermentation experiments on Panax notoginseng and Cordyceps sinensis were carried out, the main results are as follows:1.After repeated parallel experiments, with the biomass of mycelia growth a s a comparison index, and finally the wort was tested to be the right medium, and the desirable culture conditions were as pH4-5, 160 r/min, and 25℃.2.With wort as the medium, and 10% of Cordyceps sinensis and 2.5% of Panax notoginseng callus inoculation were added, and then the biomass of fermentation products was detected at the designated period of bi-directional fermentation. The results showed that it continued to increase in the range of 0-18 d, and which was mainly attributed to the growth of strain; and it reached a maximum at the 15 t h day of culture, however, it still kept increasing, the growth rate began to decline.3.The analysis of polysaccharide of bidirectional fermentation product indicated that the content of polysaccharide remained rising continuously in the range of 0-18 d, and a maximum of 5.411 mg/mL was achieved at the 18 t h d of culture; however, it decreased slowly over time of culture in the range of 18-30 d.4.Through detection of saponins of bi-directional fermentation product, it was demonstrated that the content of saponin was relatively low during the whole period of culture, compared to that of polysaccharide.5.On the base of above results, a scale- up experiment of 5L was carried out with Erlenmeyer flask. The culture parameters were set up to be 3 L of liquid media,25 C, and ventilation, and the changes in polysaccharide, protein and p H were checked every 7 days. The results showed that the content of polysaccharide was 13.04 mg/mL, 0.292 mg/mL for protein content, and the p H value was 4.51.The liquid bi-directional fermentation of Panax notoginseng and Cordyceps sinensis, aims to combine the advantage of liquid fermentation and bi-directional fermentation, set up a stable and high yielding system. It lays a foundation for the bi-fermentation of Panax notoginseng.
