Triterpenoids glycoside and its saponins were active ingredients in momordica charantia, researches showed that bitter triterpenoid compounds posseced the effect of improving insulin resistance, blood sugar, boost immunity, antibacterial. The project was separation and purification of triterpenoid compounds from bitter melon vines, and established a rapid separating methods for 3,7,25- Trihydroxycucurbita-5,23-dien-19- al(compound 1) and momordicines I(compound 2).In this paper, using several chromatography technologies for separation and purification triterpenoid compounds from 75% ethanol-water extraction, 6 known compounds were isolated and identified as 3,7,25-Trihydroxycucurbita-5,23-dien-19-al(1), Momordicines I(2), Momordicoside L(3), Momordicine IV(4), Momordicine II(5), 3,7-dihydroxycucurbita-23(R)-ethoxy-5,24-dien-19-al(6).Resin HPD200 A was screened from 7 macroporous resins by static adsorption and desorption test, and studied the mechanism of the resin adsorbed compound 1 and compound 2, the results showed that pseudo first order absorption dynamic model was suitable for describing the mechanism,and the adsorption rate was determined by membrane diffusion and particle’s internal diffusion. Then studied the adsorption and desorption properties of compound 1 and compound 2 on the resin of HPD200 A on dynamic adsorption and desorption test. The results revealed the optimization of the separation process of compound 1 and compound 2 on HPD200 A resin as follows:Adsorption: the concentration of compound 1 and compound 2 in sample solution were 0.068mg/ml, 0.091mg/ml, flow rate of sample solution was 3.5ml/min, the volume of sample solution was 3 times of the resin volume.Desorption: elution solvent distilled water(2 times of resin volume), ethanol-water solution(4 times of resin volume, 60:40,v/v), ethanol-water solution(8 times of resin volume, 85:15,v/v). the purity of the compounds in crude sample were much higher than that in sample solution, the content of compound 1 was increased 16.3-fold from 1.19% to 19.44%, while compound 2 was increased 13.7-fold from 1.60% to 21.91%, respectively. The crude sample was purified by preparative liquid chromatography, 5.3mg compound 1 and 7.1mg compound 2 were obtained,the purity was identified above 85% by HPLC.
