The function of plant growth regulator(PGR) plays an important role in the whole process of growing. During the growth of bean sprouts, the speed of growth can be promoted with plant growth regulator, but high residual of it can cause people serious harm. Many incidents of excessive levels of plant growth regulator in bean sprouts had been exposed recently, so safety issue about bean sprouts had been gained attention. In allusion to the problems mentioned above, this study as shown below established two methods for PGR testing :1. A novel simple,fast and efficient Ultra Performance Liquid Chromatography(UPLC) method was developed and validated for the separation and quantitative determination of four kinds of PGRs in bean sprouts. After extracted by methanol, the filtrate was purified by the Oasis HLB column of solid-phase extraction,the target compound in sample was monitored by UPLC. The results showed the limit of detection of 6-BA, 2,4-D, 4-CPA, NAA were 0.01 mg/kg, 0.02 mg/kg, 0.02 mg/kg, 0.01 mg/kg, the recovery values were in the range of 87.0-95.5%, and the relative standard deviation(RSD) ranged from 0.49% to 0.95% with each experiment being repeated seven times The linearity range of proposed method was 0.05-50 mg/kg(r≥0.9992). The method showed the benefits of high sensitivity, accuracy and repeatability. It makes up shortages of the existing standards that one method can monitor a kind of PGR merely and it can be used as national standard method for quality supervise department.2. A novel simple, fast and efficient Ultra Performance Convergence Chromatography(UPC2) method was developed and validated for the separation and quantitative determination of seven kinds of PGRs in bean sprouts. The results showed the limit of detection of 6-BA, 2,4-D, 4-CPA, NAA, IAA, IBA, GA were ranged from 0.001 mg/kg to 0.003 mg/kg. The recovery values were in the range of 90.7-95.5%, and relative standard deviation(RSD) ranging from 1.24% to 4.49% with each experiment being repeated seven times The linearity range of proposed method was 0.3-50 mg/kg(r≥0.999). The advantage of the method is that it has high sensitivity, accuracy, repeatability, being used less organic solvent, and it is good for the environment. The method showed potential for being used to monitor PGRs in bean sprouts.3.The change of 6-BA content with the changes of time and temperature in the storage condition of different temperatures were investigated. The results showed that the contents of 6-BA in bean sprout decreased with the passing of time. Under frozen and refrigerated, the minimum half life were 1.642 d and 3.675 d.
