Hericium erinaceus has been paid high attention by chemical and biological researchers as its edible and medical function. A large number of chemical constituents with novel skeleton were found in this higher fungus, some of which possessing significant bioactivities, such as enhancing immunity, anti-inflammatory, antineoplasmic activity and treatment of hyperlipidemia. A large aggregations of studies were concentrating their interest on the polysaccharides, and more than 80 compounds were extracted and identified from the fungus,however, there are lack of systematic researches on small molecules and their bioactivity,therefore it possesses tremendous potency.In this study, we took the mycelia fermented liquid of Hericium erinaceus as the object,examined 15 different media in order to identify the optimum fermentation condition. After incubation at 28 °C for several days, the component and content secondary metabolite were analyzed by HPLC. As a result, PD medium and rice medium were found to be the best fermentation media, and the most suitable cultivation time were 28 days and 35 days,respectively. Methanol extract of fermentation products(on large scale) was tested for the enhancement of NGF-induced neutrite outgrowth in PC-12 cells. Results showed that component of methanol phase can promote the activity and neurite length accompanied with nerve growth factor( NGF).Eight compounds were extracted from the organic extracts(ethyl acetate phase) of Hericium erinaceus, and their structures were characterized by 1H, 13 C NMR and ESI-MS. Ultimately,six compounds have been confirmed as 4-chloro-3,5-dimethoxybenzyl, Erinacine A, Bispenol A, Erinacine Q,2-hydroxymethy-1-5-α-hydroxy-ethyl-γ-pyranone and 6-methyl-2, 5-bihydroxymethy-1-5-α-hydroxy-ethyl-γ-pyranone). Two of them have been ensured as new compounds.
