Recently abalone aquaculture developed rapidly, a large number about 1/5 of the by-product of abalone in weight was abandoned, which not only caused environmental pollution, but also resulted in a tremendous waste of resources. The by-products contain a lot of oil, polysaccharide, protein and other nutrients, and those bioactive substances have very high reusable value. Considering that there are few studies on abalone viscera of steroidal compounds, the research object of this paper was about abalone viscera, which the extraction and separation process of the abalone viscera sterols was discussed; The PEF technology and the Soxhlet extraction method of extracting oil in effect on sterol yield were compared; And the sterol samples were analyzed and identified. The content of cholesterol was measured by HPLC method. The main research results were as follows:(1) The extraction process of abalone viscera oil:the process parameters of PEF on oil extraction were studied, then the best scope of each parameter was determined by single factor analysis. The optimal technical parameters determined by orthogonal analysis were as follows: PEF strength was 30kV/cm, pulse number was 8, and pulse frequency was 2700Hz.(2) The extraction and separation process of abalone viscera sterol:using the method of saponification and recrystallization to extract abalone viscera sterol from abalone visceral oil, the abalone visceral oil was saponified with KOH-ethanol solution, then extracted with petroleum ether and concentrated by rotary evaporation, finally the crude sterol was dried and obtained. At last, refined sterol was got from the recrystallization with ethanol. The parameters of saponification process were as follows:liquid ratio was 40:1, reaction time was 3h, the saponification temperature was 75 ℃, and the amount of extractant was 110mL, extracting one time. The parameters of the crystallization process were as follows:liquid ratio was 15:1, the crystallization temperature was 4℃, and the crystallization time was 24h.Different extraction methods of abalone visceral oil were compared in sterol yield. The result proved that PEF technology extracted more abalone visceral sterol than Soxhlet extraction method, which further verify the feasibility of PEF technology in extracting the active substance.(3)By analyzing and identifying the abalone viscera sterol, the structure of the compound were determined. The abalone visceral sterol samples were preliminarily identified with chemical reagents chromogenic method. It was initially viewed that the samples contain sterols which were in line with the characteristics of the color. Sterol samples were detected by GC-MS. It was confirmed that the sterol was cholesterol after compared with standard spectral library. The content of abalone visceral cholesterol was analyzed by HPLC methed which is simple,with a short peak time, and the precision experiments and recovery experiments also showed satisfactory. The result of cholesterol content indicated that 5 mL abalone visceral fats contain 176.9mg the cholesterol.
