Diosgenin is rich in Dioscorea zingiberensis C.H.Wright, mainly in the form of dioscin with cellulose exists in the cell wall, namely, by saponins in C3 and sugar chains are linked together, thus closely with plant cell walls. However, traditional methods is hard to destroy the cell wall structure which restricted the extraction yield of dioscin. And the use of acid,energy consumption and waste acid solution of hydrolysis is large amount. A novel method of stepwise biocatalysis-foam separation-preparative high performance liquid chromatography to extract dioscin of dioscorea zingiberensis. C. H. Wright has been proposed in this research. Specific research work contents are as follows:(1) Application of stepwise biocatalysis in extraction of Dioscorea sapogenins was studied. The starch and cellulose in yam were completely hydrolyzed and easily separated for the extraction of dioscin. Technical parameters of the stepwise biocatalysis extraction by the orthogonal test were determined, with the optimal parameter for extraction as hydrolysis temperature 50 ℃, p H of solution 6.0, time of enzymatic hydrolysis 2h and amount of enzymatic hydrolysis Cellulase and pectinase 50 mg. Results showed that diosgenin extraction ratio of such method was 3.30%; Also, the optimization conditions as hydrolysis temperature 65℃, p H of solution 6.0, time of enzymatic hydrolysis 4h and amount of amylase 100 mg, and extraction ratio was 3.47%.According to the step enzymatic hydrolysis optimum experiment conditions for 5 times repeated trials, yam saponin yield 3.66% ~ 3.89%. Compared with no stepwise biocatalysis, the extraction yield increased 1.37% ~ 1.60%.(2) A novel method for separating Dioscorea zingiberensis C.H. Wright Dioscin by foam separation was established in this text, based on the enzymolysis of Dioscorea zingiberensis C.H. Wright Dioscin and ultrasonic-extraction in alcoho-water. To recovery and enrichment ratio of dioscin content index. The technology of the foam isolated Dioscorea zingiberensis C.H. Wright from dioscin recovery and enrichment impact set ratio was optimized by using the single factor experiment and response surface methodology to examine the effects of variables on the extraction yields and enrichment ratio, such as dilution(concentration), temperature and loading liquid volume, the optimum condition was as follows, dilution times of 84, temperature of 20℃, loading liquid volume of 436 m L. Under this condition, enrichment ratio and recovery of dioscin reach highest to 7.53 and 91.08%.(3) After the foam separation,the concentrated foam was hydrolysised by acid, extract dioscin by petroleum ether and then recover it. Finally with ethanol to dissolve dioscin.The effects of hydrochloric acid concentration, reaction temperature, reaction time and their interactions had been investigated using single-factor experiments. The optimum hydrolysis conditions were as follows: hydrochloric acid concentration 2.5mol/Lreaction temperature100℃, reaction time 1.5h. By the analytical column linear amplification, establish efficient preparation of liquid chromatographic conditions.(4) The products obtained were characterized by GC-MS and 1H-NMR techniques.
