With the rapid development of the road transportation in our country, the coverage rate of national road network and highway mileage are also growing rapidly. The relevant designers continue to explore and study, and it is being perfected of road alignment under the process of awareness and understanding. By the natural environment as well as its high altitude special geographical conditions and other complex factors limit, the plateau highway alignment structure should have a is a big difference with the other highway,such as the elevation changes, high altitude road had to overcome a continuous longitudinal elevation; in or der to adapt the terrain, the number of longitudinal set more and longitudinal slope length and grade change frequently. Therefore, it is important to carry out research of plateau highway linear structure, designed to improve and enrich the linear theory, reduce the accident rate of plateau transportation, and improve service and road safety significantly.Based on the previous experience, the design concept of the highway always concentrates on the driving theory. Only a few people will try to design highway routes in the perspective of people and consider the human physiological property and the psychological need.Therefore, it is significant to study the physiological and psychological reaction and changing trend combined with the plateau highway’s longitudinal gradient route when people optimize the highway alignm ent design theory, improve the plateau highway’s road performance and diversify the key parameters in the designing of the plateau highway’s longitudinal gradient route. And it is also helpful to relieve or remove the driver’s feeling of tension and enhance the traffic safety.In this paper, elevated slopes of a highway at an altitude of 3,000 meters and above and the drivers driving on the slopes are used to the study for a lot of field driving experiments.The main factor of driver’s heart rate change was determined by partial correlation analysis using the binary regression growth model.The result shows that the the driver’s heart rate change increases with the rising of the elevation, and the growth rate of heart rate accerlates from an altitude of approximately 3,600 meters to higher altitudes. The driver’s heart rate change also increases with the increasing of the slope gradient. The increasing of both uphill(positive) gradient and downhill(negative) gradient gave rise to the growth rate of heart rate; The driver’s heart rate changed more distinctly under the joint effect of the slope elevation and the gradient. The higher altitude and the greater gradient of a slope, the greater growth rate of the heart rate will be.
