Cereal cyst nematode (CCN) is an important pathogens on cereal crops, a serious threat to the yield and quality of wheat in the main wheat producing areas of China. This study’s aim is to investigate the distribution of cereal cyst nematode in Hubei Province; using molecule technology of restriction fragment length polymorphism (RFLP) to analyze the internal transcribed spacer of ribosomal DNA (rDNA-ITS) of cereal cyst nematode,which is from the town of Niushou in the City of Xiangyang; using molecule technology of random amplified polymorphic DNA (RAPD) to analyze the Genetic diversity of cereal cyst nematode from the town of Niushou in the City of Xiangyang; using the dyeing method of the sodium hypochlorite-acid fuchsin to investigate the cereal cyst nematode’s life cycle. The main results are as follows:1. Currently, the survey found that the distribution places of CCN include the City of Xiangyang, Zhongxiang, Tianmen, Hanchuan, Qianjiang, it’s great amount of CCN in the town of Niushou in the City of Xiangyang, in average, it’s contains 20.61 cysts per 100ml of soil samples, and in average one cyst contains the full eggs are 140.31.The amount of CCN of others places are few.2. Digesting the area of rDNA-ITS of 10 samples of CCN groups with 7 restriction enzymes Alu I, Rsa I, Hinf I, Pst I, Taq I, Cfo I (Hha I) and Trul I (Mse I, Tru9 I), the CCN groups from the town of Niushou. We found that the CCN groups from the town of Niushou are belong to the "C" type by using the restriction map.3.20 RAPD primers,steady amplification primers,can amplify clear bands. Analyzing the genetic diversity of 20 CCN samples from the town of Niushou in the City of Xiangyang.The results shows that, RAPD bands polymorphic was 82.98%, with rich genetic diversity; the cluster analysis using NTSYS software showed that it’s more appropriate to compartmentalize the CCN groups from the town of Niushou in the City of Xiangyang with the similarity coefficient of 0.65, it’s can divided 20 CCN samples into 2 genetic lineages.4. Using the dyeing method of the sodium hypochlorite-acid fuchsin to investigate the CCN’s growthful forms and population dynamic in the root of wheat, the results reveal that the intrusion of second-stage juveniles reach the peak,while at this time the wheat is tillering stage and back green stage; at the tasseling age,there are white females exposed on the root;at the flowering stage, the amout of white females is maximum.The whole time of the life cycle of CCN is about 5 months. This study provides a theoretical basis for disease prevention, and clues for the further researchers.
