Preliminary Study Ontherelationof Viruus DsRNA And Physiology Of Pleurotus Ostreatus

Recently,the researches about the virus of Pleurotus ostreatus are emphasized.As the reports go,the rates of virus-infected mycelium grow slowly and the fruitbodies are malformal.Howerer,there are other studies which state the virus have no effect on the Pleurotus ostreatus.Qur study is mainly about virus ,which is to explain the role of virus in physiological of Pleurotus ostreatus.The results are as the followings:(1) The obtaination of virus-infected Pleurotus ostreatus strainWe selected two strains from a farm in henan,huixian.One is normal,and the other is malformal.At last,the pure mycelium were got through tissuer isolation.There are no growing differencies between the two strains.(2) The extraction of virus dsRNA by the technology of dsRNAAdd 3g grinded mycelium,we extrat dsRNA of the two strains by using the technology of dsRNA . Through 0.75%agorose electrophoresising, the result show that there are 5RNA bands in normal strain ,while 4 dsRNA bands in malformal one. The 5 dsRNA bands in normal strain include 8.2,2.6,2.4,2.1,1.1Kb,and the bands 8.2 is doublet.The malformal strain contain 4 bands,less 2.4Kb band than the normal one.It show that there are virus dsRNA in healthy mushroom and one more than the malformal’.(3) The research on the rivus elimination of normal strainThe virus-elimination teachnology of regeneration of protoplasts.0.6%Mannitol is the Osmotic stabilizer.We dissolve the mycelium using 1.6% wall-solution enzyme for 3 hours at 30℃, getting 20 regeneration protoplasts.We choose 7strains which contain different dsRNA bands.There is only one strain without dsRNA in 20 regenerations,and the other are all virus-containing strains,which are not eliminated completely.2.4Kb bands are all eliminated in all selected 20 regenerations.It indicate that 2.4kb band is not coated by capsid protein and maybe 2.4Kb is benefit to formation of fruiting. (4) The research on Physiological Characteristics of 8 strainsWe compare the 8 strains ,finding that the strain containing most dsRNA is more than orther s in rates of mycelium ,production and enzyme activity .We also find that the prime strain is freeze-resistence.On the contrary,the strain without dsRNA is inferior to the ones containing dsRNAs,whose enzyme activity are mostly the lowest .It indicates that the viruses have no bad effects on the Pleurotus ostreatus.The existing virus is not sure to make the mushroom malformal,to the contrary,it has good effects on the growth of Pleurotus ostreatus.(5) Sequencing of band 2.4Kb by single primer amplification technique. After sequencing the segment of 2.4Kb, Sequences obtained were compared with the known sequences in GenBank,and there were no identical sequence with the 2.4Kb.It indicate that 2.4Kb is a new virus band or only a dsRNA in Pleurotus ostreatus.
