| 85 pleurotus ostreatus strains were detected by the dsRNA technique. The result revealed that 34 strains have dsRNA virus.They were 40 percent in all detection strains, so dsRNA virus existed widespreadly in pleurotus ostreatus. According to electrophoresis map, the dsRNA virus had four segments those were 8.0kb, 2.3kb, 2.0kb and 1.0kb in size, respectively(according to the calculation Bi0CaPtMW software). According to the different of dsRNA segments, they could be roughly divided into five categories.According to the known sequence of pleurotus ostreatus virus in Genebank, 10 pairs of primers were designed corresponding the three larger dsRNA segments. Finally, we searched for 1 pair of primers respectively. cDNA were obtained by reverse transcription (RT-PCR), clone and sequenced. By blast comparison analysis, it proved preliminary that the dsRNA viruses those we extracted were the same as the virus extracted from pleurotus ostreatus 300 by Qiu Liyou.The dsRNA virus were eliminated 4 generations continuously by virus elimination technique of hypha tip.We got some virus-eliminated strains. DsRNA segments obviously weaken or reduce by the dsRNA technique.It suggested that the amount of virus and genome reduced.The average of hypha growth speed of virus-eliminated strains improved 28.1 percent. Carboxy methyl cellulose enzyme activity and Laccase activity improved 33.3 percent and 20.6 percent respectively.Yield increased 13.6 percent, and the ability to fight pollution also improved.The vertical transmission rate of dsRNA virus through spores was 7.7 percent. So the transmission of dsRNAs via basidiospores is very inefficient in pleurotus ostreatus. By Buhler hybridization and the single-single hybridization, the horizontal transmission of dsRNA virus was researched.The result suggested that dsRNA virus can transmit between compatible strains with virus-infected and virus-free. |
