The conclusions are as follows:1. The determination of leaf surface trichome exudate and carotenoids was higher in the new flue-cured tobacco varieties 8306 and Y892, and the high determination of leaf surface trichome exudate was the remarkable characteristic of 8306, and the high leaf determination of leaf carotenoids was the remarkable characteristic of Y892. Moreover, The determination of leaf surface trichome exudate of filial generation Y892(K326♀ⅹ8306♂)was between its parents, and proved that inheritance of leaf surface trichome exudate was quantitative trait, it’s beneficial to improving the determination of leaf surface trichome exudate of filial generation when explained that the male parent of higher determination of leaf surface trichome exudate and the male parent of lower determination of leaf surface trichome exudate hybrid. And inheritance of leaf carotenoids has heterosis, The determination of leaf carotenoids of filial generation Y892 was higher than its parents.The differential expression of key enzyme genes involved in upper terpenoid biosynthetic pathway in different flue-cured tobacco was studied. The results showed that, the expression of ipi play an important role in accumulation of terpenoid, but it always accompany the other genes’high expression, and proved that the accumulation of terpenoid was the result of multiple genes, and only depend the expression of ipi is difficult to stability and improve the determination of terpenoid. The expression of key enzyme genes involved in lower carotenoid biosynthetic pathway in different flue-cured tobacco was different from each other, these genes decides the components and its determination of carotenoids and are also the molecular basis of forming the special tobacco.2. The determination of leaf surface trichome exudate and carotenoids were significantly different on 20 and 60 leaf age among the genotypes 8306, Y892, tobacco 100 and K326, the ordinary nitrogen treatment was significantly higher than the high nitrogen treatment and low nitrogen treatment. The differential expression of 8 key enzyme genes involved in the upper terpenoid biosynthetic pathway under the regulation of different nitrogen levels was studied. The results showed that, 7 of the 8 key enzyme genes’expression of low nitrogen treatment were significantly higher than the high nitrogen treatment and ordinary nitrogen treatment in the 4 flue-cured tobaccos on 20 leaf age. And it is considered that the highest expression of the key enzyme genes didn’t cause the content of leaf secretions and carotenoids increased under low nitrogen treatment is associated with the growth of tobacco slowdown caused by nutrition deficiency, because terpenoid metabolism is closely related to hormone synthesis (GA and ABA) of tobacco. There were 8 and 6 tobacco 100 and Y892 was the highest expression under low nitrogen treatment, 6 and 8 genes separately of K326 and 8306 was the highest expression under ordinary nitrogen treatment on 60 leaf age. But the content of leaf secretions and carotenoids were highest under ordinary nitrogen treatment, and this is associated with the maturity degree and the changes of hormone content in different genotypes tobacco under different nitrogen treatment.The results of the differential expression of 6 key enzyme genes involved in the lower terpenoid biosynthetic pathway under the regulation of different nitrogen levels showed that, the expression of 6 key enzyme genes involved in lower carotenoid biosynthetic pathway in different flue-cured tobacco under the same nitrogen level and the same tobacco under different nitrogen levels were different from each other, and it showed that the rate of nitrogen can regulate the expression of key enzyme genes which control the components and its determination of the leaf carotenoids.3. In the treatment of salicylic acid, the determination of leaf surface trichome exudate of the three varieties of flue-cured tobacco(8306, Y892, Zhongyan 100) have little effect on two leaf ages, and The determination of leaf carotenoids have little effect on 25 leaf age, and increased on 45leaf age. And it showed that SA is little effect on the determination of leaf surface trichome exudate, but it can increase the determination of leaf carotenoids.In the treatment of salicylic acid, 8 out of 11 11 key enzyme genes involved in terpenoid biosynthetic pathway in the three varieties of flue-cured tobacco had the same change tendency, suggesting that the related genes involved in isoprenoid metabolic pathway possessed the same or the similar ecological behavior in response to the treatment of salicylic acid. In the treatment of salicylic acid, the changes of determination of leaf surface trichome exudate and carotenoids of the three varieties of flue-cured tobacco has little effect on 25 leaf age, but the changes of key enzyme genes may result of autoregulation of the tobacco to environmental impact. The determination of leaf carotenoids increased and the determination of leaf surface trichome exudate has little effect on 45 leaf age, suggesting that it may be the changes of determination of other terpenoid and hormone synthesis caused the changes of components of the terpenoid. 4. The variance analysis results of the determination of leaf surface trichome exudate on K326, NC89 and Hong Hua Da Jin Yuan in different ecological conditions(Henan, Yunnan and Guizhou )showed that, the determination of leaf surface trichome exudate on K326 planted in was much higher,and the determination of leaf surface trichome exudate on NC89 planted in yunnan was much higher. the determination of leaf surface trichome exudate on K326 was the lowest during the three tobacco varieties in Three ecological regions. The determination of leaf carotenoids of the three tobacco varieties in three areas was different , Yunnan was the highest , Guizhou was the second , Henan was the lowest . The determination of leaf carotenoids on K326 was the lowest during the three tobacco varieties in Three Three ecological regions.The differential expression of key enzyme genes involved in terpenoid biosynthetic pathway of the middle leaves of general genotypes tobacco (K326, NC89, Hong Hua Da Jin Yuan) on 20, 40, and 60 leaf age in different ecological regions (Henan, Yunnan, Guizhou) was studied.
