Study On Degradation Regularity Of Cell Wall Components Of Five Typical Fibrous Feedstuff In Animals Gastrointestinal Tract

The aim of this project is to determine the degradation schema of cell wall components of typical fibrous feedstuff in animal gastrointestinal tract. Meanwhile, partition of dietary fiber into SNSP and INSP, minerals, monosaccharides constitute of different kinds of analyzed fibers residues were detected.Experiment 1 The gross nutrient contents including crude protein, detergent fiber,amino acids and minerals of five fibrous feedstuffs, i.e, Soyhulls, Alfalfa meal, Broccoli residues,Wheat bran and Rice bran were analyzed.The results showed that the contents of crude protein and amino acids were 13.46%,18.93%,16.04%,21.17%,21.07% and 12.82%,17.5%,14.2%,19.2%,16.2%, respectively. Additionally, the contents of Ca, Mg and Fe were relative higher when compared with that of other feedstuff. Also, The results revealed that the contents of NDF,ADF,CF of the five feedstuff were as follows:(1)NDF: 62.1%,24.9%,27.7%,33.8% and 20.7%;(2)ADF:49.6%,30.3%,20.5%,10.4% and 12.2%;(3) CF:37.2%,23.2%,16.7%,8.7% and 8.6%. Obviously, the content of amino acids, minerals and detergent fiber of the fibrous material from different sources is difference.Experiment 2 The method to analyze neutral sugars composition and uronic acids of dietary fiber was verified by European reference material CRM 518 Bran Breakfast Cereal using the Uppsala Method.The content of SNSP and INSP above mentioned feedstuffs was quantified as the sum of individual in soluble and insoluble part.The monosaccharides constitute of NDF,ADF and CF residues wew also analyzed. The results showed that the contents of INSP of the five feedstuff were ranged in 25%-55%,whereas the SNSP were in very low contents, only ranged 2%-4%. The ratio of monosaccharides as Gal, Ara, Xyl, Man, Glu and UA in NSP of alfalfa meal,Broccoli residues,Wheat bran and Rice bran was 10:17:23:8:49:34,10:10:15:7:76:35,10:94: 141:13:127:13 and 10:33:36:10:74:12,respectively. Further,the ratio of Glu in DF of the aforesaid five feedstuff was only 18%-45%,while Glu in the detergent fiber residues of ADF and CF in the proportion of total polysaccharide accounted for 80%-90%.In addition,the monosaccharides of Wheat bran as Gal, Ara, Xyl, Man, Glu and UA in DF and NDF in the proportion of the total polysaccharide accounted for 23.6%,35.2%, 31.5% and 24.9%,35.8%,33.4%, respectively.While in ADF, Ara, Xyl and Glu accounted for only 0.4%,9.4%,84.6% and CF of 0.6%,8.5%,87.6%, the similar as rice bran. The total contents of monosaccharides in NDF and ADF were found only significant higher than that of CF in Alfalfa meal and Broccoli residues (P<0.05).And the monosaccharides were all significant different in NDF,ADF and CF when compared that with each other(P<0.05) in soyhulls,wheat bran and rice bran. In summary, there was a phenomenon showed that the content of UAs was only higher in Soyhulls,Alfalfa meal and Broccoli residues whereas the contents of Xylose(Xyl) and Arabinose (Ara) were higher in wheat bran and rice bran. Besides, Glucose(Glu)was found to be the highest monosaccharide in all these five feedstuff.Experiment 3 Thrity adult egg-type breed cockerels with similar age and body weight were randomly allotted into five groups, six birds in each one. Each bird was feeding in separated cage individually as a replicate. Five semi-purifine diets were organized using one of the above feedstuff as sole cell wall origin in each diet. Each group of birds were randomly assigned one diet by force feeding method.The chyme of crop,stomach, small intestine, ileum and cecum were taken after killing the adult cock. To analysis the contents and changes of minerals and monosaccharides in diet, small intestine,ileal and fecal samples and the relationship of them.The results showed that the apparent digestibility of soluble monosaccharides and UAs in the five diets were ranged 40%-65%,and the apparent digestibility of dry matter(DM)and ether extract(EE)were the highest in rice bran with the value of 50% and 80%,respectively.The contents of NDF,ADF and CF were found decreased in the whole GIT of cockerels, with a tendency of increase only in the hindgut. A big difference was observed between NDF and ADF in diets of wheat bran and rice bran. Additionally, an obvious tendency of decrease of SNSP in the whole GIT was found in the five diets, and an obvious inverse correlation between SNSP and minerals was also observed. In contrast, there was little change of INSP in the whole GIT of cockerels, and the inverse correlation between INSP and phosphorus(P) was also observed in this study.
