| Hanzhong is located in the southwest of shaanxi province, its north is Mts. Qingling andits south is Mts. Bashan. The climate of Hanzhong is mild, with abundant precipitation, thetemperature between day and night is different obviously. HanZhong is on the northen edge ofthe most appropriate growing areas of citrus in China. The citrus industry has become a pillarindustry of agricultural development in HanZhong city. The Citrus germplasm resourcessituation, tasting and processing quality analysis study are important in its fruit breeding andvariety extension, cultivation technology research and base construction, citrus storage andprocessing, product quality evaluation as well as industrial development.Now, citrus germplasm resources, taste and processing quality analysis system inHanZhong is still lack of research work. This study focused on the three aspects:(i) The citrusgermplasm investigated and analysed,(ii) the eating quality evaluated and (iii) its processingquality evaluated. The results of this study were listd as followings:(1) The Citrus germplasm resources in Hanzhong were completed by ways ofinvestigation, literature retrieval, related units visitting, discussion and the field survey. As theresults showed, two antiquity citrus, Rough-peel Mandarin (before Tang Dynasty) andZhonghongju (before Ming Dynasty), had been planted before1949year. More than80citrusvarieties were imported after1980. Then, only27citrus varieties were knocked out throughscreened. According to their botany characteristics, they were sorted as Citrus grandis,Aurantium and Citrus reticulata. Citrus reticulata had23varieties, with85.2%of the citruscounted. Both Aurantium and Citrus grandis had2varieties, with7.4%of the citruscounted. The main varieties in Hanzhong were Fanyahuanghou, Yamasaki,Chenggubingtang, Nichinan1, Wakiyame, Okitsu, Fanyabingtangju, Fanyahongmi,Zhuhongju, Shatian pummelo, Fanyaxiangcheng, Ooura, Miyagawa with the40.7%ofthe citrus counted. The Okitsu and Miyagawa accounted for95%of the cultivated area.(2) The comprehensive evaluation and the eating quality of cultivated13main cultivated citrus of HanZhong were carried out by sensory evaluation and index evaluation method. Theresults showed the different varieties citrus had difference eating quality. The fruit type indexwas0.63-1.14.The Yamasaki has good shape. The fruit type uniformity coefficient was14.48%and Yamasaki was uniformity. All the citrus had fragrant flavour, sour and swee.Thetotal sugar content was69.08-104.65g/kg. The soluble solids content was10.25-13.48%. Thetotal acid was8.66-19.05g/kg. The sugar acid rate was4.28-11.28. Fanyahuanghou, Yamasaki,Chenggubingtang, Nichinan1and Wakiyame were fine eating quality.(3) AAS, HPLC, UPLC-MS/MS and the fuzzy membership function were imployed toevaluate the processing quality of the main cultivated citrus of Hanzhong. As the result,different varieties citrus had different processing quality. The trace elements were rich incitrus. The content of heavy metals fitted to the People’s Republic of food sanitation standard.The content of vitamin C was244.34-392.92mg/kg. The content of limonin was3.87-80.78mg/kg. The content of nomilin was1.0-12.61mg/kg. The content of naringin was230.71-637.71mg/kg. The juice rate was in32.35-52.27%.The skin rate was in14.29-28.50%.Nichinan1, Fanyahuanghou, Yamasaki, Fanyabingtangju, Chenggubingtang were offine processing quality.(4) A UPLC-MS/MS method to analysis limonin in citrus were established. The columnwas waters ACQUITY BEH C18(50mm x2.5mm,1.9μm). Mobile phase: Using gradientelution, A was acetonitrile and B was water, A:10%(0min)-50%(3min)-10%(4min)-10%(5min).The velocity of flow was0.3ml·min-1.Column temperature was40℃.Injection quantity was5μL.Detection time was5min. ESI (+) scan, the limonin’s parents m/zwere471.1, daughters m/z were425.3and m/z=161.1.The separating degree was R>1.5,recovery was100.67%(RSD=2.08%, n=5), repeatability RSD was1.99%(n=5), stabilityRSD was2.30%, detection limit was2.8ng/mL. This method can satisfy the citrus fruitquality analysis requirements, and be used for limonin analysis in citrus. |
