In this research the citrus reticulata blanco varas was used as test material,through the mature stem segments of in vitro culture emerge the shoot meristems,preliminary analysis the seedling in vitro technology with stem in mature citrusreticulata blanco var. Discuss in which grafting way and culture conditions have thehighest survival rate,then analysis the genetic stability of grafted seedlings, throughcompare with the graft seedlings and cultured in vitro citrus reticulata blanco varvarious of isozyme similarities and differences,thus this study provide certaintheoretical and technological foundation to citrus reticulata blanco var rapidpropagation system.And the paper results are as follows.1. Through citrus reticulata blanco var tissue culture induced adventitious buds,the research shows that MS+6-BA0.3mg/L+ZT0.5mg/L+NAA0.05mg/L+folicacid0.1mg/L+sucrose50g/L have the highest rate in produce adventitious buds,added ZT and increase sucrose concentration can increased sprout rate significantly.2. The tube grafting technology test shows that the seeds must be selected singleembryo, which the seedling stem thick and strong,thus can convenient operation,moreover the seeds leaves should be few that can reduce side of the leaves growthrestrain graft union process.3. In this study used side grafting method can avoid top grafting loss water andfalls off phenomenon, and also can prevent T grafting and the T glyph grafting callusserious phenomenon in the incision.4. Experimental shows that the10d growth period seed have the highest graftsurvival rate, young and half lignification stock in operating can reduce the hurt, thetoo long and too short seeding disadvantage the graft process.5. The experimental research shows that4leaf primordium size have thehighest survival rate, if the stem tip too small to loss water, too big to fell from thestock.6. Training mode is also an important factors influence the graft survival rate,in this research using MS+GA1mg/L+75g/L sugar concentration have the highest graft survival rate, and found first dark cultivate a week then into the light train canreduce scion loss water, in this way can improved the graft survival rates.7.The research shows that seeds in different transplanted matrixs have nosignificant difference, all the transplanted matrixs survival rates more than90%.Analysised the various of seedings isozymes, research shows that the seedingsand the mature of adult citrus reticulata blanco var cultured root buds in the POD、CAT and SOD isozyme molecular expression have no obvious difference, andmorphologic observation aslo have no obvious difference.Thus we can draw aconclusion that through the shoot mico-grafting technology can keep the femaleisozyme molecular expression.
