In this research, seeding and mature stems in vitro culture was studied, and has established a set of practical and effective regeneration system. Different physiological and biochemical mechanism was studied on the endogenous hormones, the activity of POD, POD isozyme analysis. Results:1.This paper, the cotyledon, epicotyl, stick, hypocotyls Citrus reticulate blanco were used as explants for buds and roots on culture media with different hormone combinations. The best results were achieved on the following media: (1)90% shoot induction, the best part is epicotyl; (2)The best concentration media is MS+6-BA 0.8mg/L+NAA0.1mg/L+D-calcium d-pantothenate 0.5mg/L in the subculture medium;(3) rooting on 1/2MSmedium supplement with NAA0.1 mg/L.2. In vitro culture of mature stem, indoor varieties have lower rate of pollution than the wild varieties, materials of 0.8cm stem section has lower rate of pollution in autumn.3.By studied on mature stems in vitro culture, obtained internode shoots and get a few varieties of initial medium formulation."YangXiao2-6"mature stems in the MS+6-BA0.8mg/L+IAA0.2mg/L+LH500mg/L+folic acid 0.1mg/ L medium,was the best induction effect, bud germination rate wsa 82% and the number of induced axillary bud more growing strong: In the MS+6-BA0.4mg/L+IAA0.2mg/L+LH500 mg/L+folic acid 0.1 mg/L medium and"small fruit SS-28"mature stems of the axillary bud induction was the best: MS+6-BA0.8mg/L+IAA0.4mg/L+LH500mg/L +folic acid 0.1 mg/L medium was best suited to"small fruit SS-7"and"large fruit 97-1".4.Endogenous phytohormone levels and the balance played an important regulating role in Plant regeneration in vitro de-differentiation, regeneration, and plant growth. By measuring the changes in endogenous phytohormone of in vitro plant regeneration, showed high ZT benefit the production of adventitious buds, GA could promote the growth of buds, ABA could inhibit plant growth. While high levels of growth hormone was main reasons of mature stem segments formed calli. The study also showed that the interaction of endogenous phytohormones is mutual, not a single of plant regeneration.5. By the determination of POD activity and separation of POD isozyme, we can find a certain variation of POD as the train growth stage, and the analysis of the POD isozyme, can find the POD isozyme with depth, width and enzyme amount show certain differences. They could be used to analyze the physiological and biochemical indexes in the process of cultivation.
