This experiment was conducted to study the effect of submerged fermentation concentrate ofHericium caput-medusae (Bull.:Fr.)Pers. on the cecum’ Lactobacillus in broilers. A total of300one-day-old AA broilers were divided randomly into5groups with3replicates of20broilerseach. The control group was fed by the based diets, the antibiotic group was fed by the baseddiets with0.5%flavomycin,the experimental groups were fed by different amount of HFC in thebased diets(concentrations of polysaccharide in the based diets was0.1%and0.3%and0.5%)during the60days study.The results as followed:1. In the HFC,the polysaccharide was18%, the water was33.08%, CP was4.68%, the adiposewas11.29%, the fiber was0.39%, the ash was1.99%, the energy was4.13MJ.Kg-1, thecontent of Ca was3.86%, the content of P was1.95%.2. The results of the production performance with broilers:in21days, the group of HFC couldincrease the average daily gain and reduce the feed conversion rat;in42days,the group ofHFC could increase the average daily gain and the average day intake,at the same timereducing the feed conversion rat;in60days, the0.1%group of HFC could increase theaverage daily gain and reduce the feed conversion rat. In a word,HFC could promote theproduction performance of broilers.3. The quantitative analysis method of real-time FQ-PCR on the Lactobacillus was established,the regressing equation Y=-3.337*LOG(X)+35.31, the relative coefficient R2=0.998, theamplification efficiency was99.4%. The sensitivity:template detection limit was47.3copies/μL; the specificity: the melting curve was single, indicating that the method wasspecific; the repeatability: the intra-assay coefficient of variation was0.31%and0.3%, theinter-assay coefficient of variation was1.21%and1.45%in the standard of4.73×107and4.73×105.4. The quantitative analysis method of real-time FQ-PCR on the Lactobacillus was established,the results showed that: in21days, the0.1%group of HFC could significantly increase theamount of Lactobacillus(P<0.05); in42days, the0.1%0.3%0.5%group of HFC couldsignificantly increase the amount of Lactobacillus(P<0.01); in60days, the0.5%group ofHFC could significantly increase the amount of Lactobacillus(P<0.05). In a word, HFCcould increase the amount of Lactobacillus.Conclusion: 1.The quantitative analysis method of real-time FQ-PCR on the Lactobacillus was established.2. HFC could promote the production performance of broilers, and the optimum supplemental levelis0.1%.3. HFC could increase the amount of Lactobacillus in the cecum of broilers, and the optimumsupplemental level is0.1%.
