Panax quinquefolius L. is also known as American Ginseng. It’s perennial herb inAraliaceae ginseng. Originating in the United States and Canada. In the70s of twentieth century,the American ginseng was introduced successfully to China, and it has become one of importantspecies of ginseng. Modern pharmacology enhance that American ginseng can increase immunefunction, anti-fatigue, anti-hypoxia and myocardial protection, as well as anti-arrhythmia,anti-atherosclerosis and decrease blood sugar. The main active ingredients of ginseng isginsenoside. Ginsenoside become a focus of attention of scholars for its great medicinal value.The experiment presented in this paper is based on hairy root of Panax quinquefolius L,suspension cultured the hairy root in1/2MS medium, then screened the best fortifingconcentration, adding time and acting time of SA. The results showed that when added0.001mg/L of SA at the20thday of the culture period,5days later, the biomass of hairy root increased13.74times and the ginsenoside was1.2times more than the control groups.The experiment determined the change of the activity of POD, CAT and PPO in hairy rootof Panax quinquefolius L. after the addition of SA in72hours. The results indicated that afterthe addition of SA, the activity of POD reached the highest level in24hours, was1.62timesthan the control groups; the activity of CAT was improved steadily in24hours, was1.2timesthan the control groups; the activity of CAT was improved steadily in20hours.Determining the expression’s variations of key genes in ginsenosides with adding elicitor in72hours by SYBR green I real-time qPCR technology. The results showed that: It hadsignificantly increasing the expression of enzyme genes (SQS、SQE、DS、β-AS and OSC) in20-24hours, so as to rise the synthesis of ginsenosides. It proved that adding elicitor could makea contribution to the strength of metabolic activity in hairy root cells,and conduciving tolarge-scale synthesis of secondary metabolites.This study investigated the transcription level expression’s variations of synthesis enzymegenes in ginsenosides with adding elicitor, it laid an important theoretical basis in furtherrevealing the role of genes and in-depth studying to explore the biosynthetic pathway ofginsenosides, and also providing theoretical basis and technical support in mass production ofginsenosides by taking technology of hairy root of Panax quinquefolius L.
