Eupolyphaga is a kind of traditional Chinese medicinal materials in our country,has been recorded in the ancient classic books.It can broke of blood stagnate,swelling volume and through the meridians.It is also used as a food to eat,because of its low toxicity and rich in protein.Pharmacological studies showed that Eupolyphaga can regulate blood lipid and has the effects of antitumor and oxidation,and so on. It can be used to treat and prevent disease.This experiment was to study the effects of Eupolyphaga Freeze-dried powder on blood lipid physiological indexes in rabbits with high-fat diet and the expression of related genes, explore the possible molecular mechanisms of regulating blood lipid level of Eupolyphaga Freeze-dried powder.150rabbits were selected and randomly divided into five groups,control group(normal feed),high-fat group(high-fat feed) and three different dosage Eupolyphaga groups(0.4g/kg,0.2g/kg,0.1g/kg Eupolyphaga Freeze-dried powder). Every10days,each group of randomly selected5rabbits to weight and draw cardiac venous blood, separation of serum,used to measure the serum’s total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C) levels. Test the20th and40th days,each group of randomly selected3rabbits to kill and get liver, used to study the gene expression of time and space of ApoE gene and LDL-R gene who are related with the blood lipid.Discussing the molecular mechanism of the effects of Eupolyphaga Freeze-dried powder. The main experimental results show that:1. No significant differences(p>0.05) were observed between rabbit’s weight of each group.The organ index of liver of high-fat group was abnormal, compared with control group, the levels were significantly increased(p<0.05); Compared with control group and high-fat group, the organ index of liver in high group and low group were increased(p>0.05). Compared with control group, the organ index of liver in medium group had a tendency to reduce (p>0.05). Compared with high-fat group, the organ index of liver in medium group were significantly reduced(p<0.05).2.The results showed that the blood lipid of high-fat group was abnormal, compared with control group, the levels were significantly increased(p<0.05);Compared with control group, the levels of TC^TG in high group and low group were significantly increased(p<0.05), no significant differences(p>0.05) were observed between high group,low group and high-fat group;Compared with high-fat group, the levels of LDL-C in high group,medium group and low group were significantly reduced(p<0.05), no significant differences(p>0.05) were observed between high group.medium group, low group and control group;Compared with control group and high-fat group, the levels of HDL-C in high group,medium group and low group were significantly increased(p<0.05);Compared with high-fat group, TC, TG, LDL-C of medium group were significantly reduced(p<0.05),and no significant differences(p>0.05) were observed between the medium group and control group.3.The gene expression of ApoE in livers of high-fat group was abnormal,compared with control group, the levels were significantly reduced (p<0.05); No significant differences(p>0.05) were observed between high group.low group and control group(p>0.05). No significant differences(p>0.05) were observed between high group and high-fat group(p>0.05). Compared with high-fat group, low group were significantly increased(p<0.05);Compared with high-fat group.control group, high group and low group, medium group were significantly increased(p<0.05).4.The gene expression of LDL-R in livers of high-fat group was abnormal.compared with control group, the levels were significantly reduced (p<0.05); No significant differences(p>0.05) were observed between high group,low group and control group(p>0.05); No significant differences(p>0.05) were observed between high group,contral group and high-fat group(p>0.05);Compared with contral group, high-fat group and high group, medium group were significantly increased(p<0.05). Among them, medium group was the best.Conclusion:Eupolyphaga Freeze-dried powder can inhibit the rise of TC, TG, LDL-C of blood lipid in rabbits fed with high-fat diet. Eupolyphaga Freeze-dried powder can make HDL-C significantly increased.lt can prevent some diseases such as high cholesterol and so on.The mechanism of Eupolyphaga Freeze-dried regulating blood lipid levels may be related to improve the ApoE gene and LDL-C gene expression. The best dosage of Eupolyphaga Freeze-dried preventing the rising of blood fat is the medium group (2g/kg), medication time is about40days.
