Hyperlipidemia is a disease caused by disorder of blood lipid, usually represented as blood lipid abnormality increased, is a high risk factor for atherosclerosis, coronary heart disease, cerebral infarction and other high-risk disease. Hyperlipidemia also is a common disease among people, and has a tendency toward younger. Hirudo is a traditional Chinese medicine with a long history, and can removing blood stasis and preventing epilepsy. Modern research shows that Hirudo has anticoagulation, antithrombotic, antitumor, anti-inflammatory, anti-fibrosis, etc. In clinic, Hirudo can use to treat cardio-cerebrovascular disease, diabetes, liver cancer, amenorrhea, blood hyperviscosity syndrome, nephritis, prostatitis etc. With the development of science and technology, the research methods are gradually improved and being perfected, the study of Hirudo is also gradually in depth and comprehensive.The purpose of this experiment was to study the serum lipid regulation effect and the mechanism of hyperlipemia of Hirudo in rabbits. In the experiment,150male rabbits, which average body weight was (2.045±0.031) kg and3mouth old, were randomly divided into5groups. The control group were fed with standard diet, the high lipid group were fed with high-fat diet, the other three Hirudo groups (high dosage group, middle dosage group and low dosage group) were fed with high-fat die and were daily fed with0.15,0.075,0.0375g/kg Hirudo powder at the same time for40days. In the experiment, the blood samples were drawn from heart in every5days and tested the level of total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), the low density lipoprotein cholesterol (LDL-C) was calculated by formula. In20d and40d,3rabbits in each group were killed to get liver samples and plunged into liquid nitrogen immediately, using Real-time PCR determinate the levels expression of ApoAl and ApoE in livers of rabbits. The main results are as fallows:1、At the end of experiment, compared with control group, the TC、TG、LDL-C level of high lipid group were significant increased (p<0.01), showed that after feeding with high-fat diets for40days, were successfully established hyperlipemia model. Compared with high lipid group, the TC、TG、LDL-C level of three Hirudo groups were significant decreased (p<0.05,p<0.01); Compare with control group, the HDL-C level of high dosage group significant increased (p<0.05), showed that Hirudo can reduce the serum lipid level of rabbits and prevented the formation of hyperlipemia. The TC、TG、LDL-C level between three Hirudo groups were no significant (p>0.05), showed that the effect of different Hirudo dosage were equally. Compared with low dosage group, the HDL-C level of high dosage group were significant increased (p<0.05), proved that the effect of high dosage Hirudo are better than other Hirudo groups.2, At the end of experiment, compared with control group and high lipid group, the ApoA1gene expression in liver of high and low dosage group were significant increased (p<0.05,p<0.01), showed that high and low dosage Hirudo could regulated the ApoA1gene expression in rabbit liver. The regulation effect of ApoA1gene expression of different Hirudo dosage from high to low was low dosage group, high dosage group and middle dosage group, the difference in each group was significant (p<0.05,p<0.01).3、At the end of experiment, compared with control group and high lipid group, the ApoE gene expression in liver of three Hirudo groups were significant increased (p<0.01). This result showed that different dosage Hirudo could improve the ApoE gene expression in rabbit liver. And the effect on ApoE gene expression between different dosage Hirudo was no significant (p>0.05).In summary, the result of the study suggested that Hirudo can effectively regulate the serum lipid of rabbit, reduce the serum lipid in high-fat feeding rabbits, restrain the formation and development of hyperlipemia in rabbits, prevent the occurring of hyperlipemia, atherosclerosis, coronary heart disease and other related disease. There is no significant difference between different dosage Hirudo of effect on the serum lipid level, but after considering reducing cost, making full use of medicine and protecting the resource of medicine, the best Hirudo dosage for decreasing serum lipid and preventing hyperlipemia is0.0375g/kg.
