Lonicera edulis Turcz is a wild berry resource which nutrient value is very high. There are many resources in our country, but the wild Lonicera edulis Turcz grows dispersedly, the fruit is usually very small and hard to pick, it is unable to meet the market demand. So there is an urgent need to screening out the superior individuals from the wild resource and which is needed to be artificial cultivated and promoted. Therefore, the study explored the influence factors of proliferation and rooting, and the effect of different transplanting substrates on survival rate in the process of domesticating tissue culture plant which takes the stem of superior individuals of Lonicera edulis Turcz which is selected from the wild resource of Changbai mountain as material. Then conduct the commercial production. The results showed that:l.The most suitable proliferation medium for tissue seedling of Lonicera edulis Turcz is MS+6-BA0.8mg·L-1+IBA0.1mg·L-1, and the proliferation times can be up to6.93, in which the seedlings grew strong with high proliferation. White sugar was better than sucrose as sugar source, and the most suitable concentration of sugar was30g·L-1for proliferation.2. The modified MS was the best medium for rooting in vitro, the rooting growth showed a trend of increased then dropped with the increase of concentration of IBA. When the concentration of IBA is1.0mg·L-1, the rooting rate can be up to100%. Meanwhile, the callus was less, the roots were strong and the number of the fibrous roots was most which is most suitable for the growth of Lonicera edulis Turcz.3. In the experimentation of ex vitro rooting, the most suitable concentration and treating time of the quick dipping of high concentration method was2000mg·L-1of IBA and10s, the rooting rate could reach83.33%, and the survival rate could reach93.33%. The most suitable concentration and treating time of the soaking of low concentration method was250mg·L-1of IBA and5min, the rooting rate could reach86.67%, and the survival rate was93.33%. There were no significant differences between the two methods. So in order to save time and make it easy to operate, the quick dipping of high concentration method is recommended in practical large-scale production.4. Humus:Pearlite=1:1is a relative suitable transplanting substrate for Lonicera edulis Turcz which had the best effect, the survival rate was93.33%. And the growth situation is better than other combinations of matrix.5. Sucrose can be replaced by white sugar in the commercial production of Lonicera edulis Turcz, it could greatly reduce the costs. The ex vitro rooting could replace in vitro rooting, it could save costs and shorten the culture cycle. The most suitable period to transplant domestication in the field is the middle of June for tissue culture plant of Lonicera edulis Turcz, the temperature and sunlight are suitable for growth. The survival rate is high and growth speedy; the tissue culture plant can be planted in the same year.
