This paper minly studied the extraction of volatile oil in perilla frutescens by Water distillation, and then identified the constituents. Then we studied the effect of Perilla frutescens volatile oil, perillaldehyde and limonene on growth and immune function of mice. The three experiments are as follows:Experiment1was carried out to obtain volatile oil of Perilla frutescens from dry Perilla frutescens leaves by Water distillation. The extraction condition were:4%of NaCl,1hour for immersion,5hour for distillation, and solid-liquid ratio1:6, the actual detection value was0.3677%.Gas Chromatography was used to detected perillaldehyde and limonene concentration, which was284.8mg and151.0mg per milliliter of volatile oil, respectively.Experiment2was to study the effect of the volatile oil from perilla frutescens on mice weight, blood serum index and immunne performance. Sixty mice were divided randomly into4groups (n=15for each) and orally infused to stomach with different concentration of volatile oil: high(0.04mL/kg), middle(0.03mL/kg), low(0.02mL/kg); the control with0.2%-Tween-80,250μL once daily for21days. The results showed that compared with the control group, the mice’s body weight, spleen and thymus index, the level of macrophages phosphatase activity (ACP) and IL-2in high and middle groups were extremely significantly increased (P<0.01) the level of IgM and IgG in high and middle groups were significantly increased (P<0.05).The number of Escherichia coli in high and middle groups were extremely decreased (P<0.01) Experiment3was to study the effect of Perilla frutescens volatile oil, perillaldehyde and limonene on growth and immune function of mice. Sixty mice were divided randomly into4(n=15for each) groups and orally infused to stomach with control(0.2%-Tween-80), Perilla frutescens volatile oil(0.03mL/kg), perillaldehyde(8.54mg/kg) and limonene(4.53mg/kg);250μL once daily for21days. The results showed that compared with the control group,(1) in volatile oil group, the number of Escherichia coli were extremely decreased(P<0.01), and blood urea nitrogen were decreased significantly(P<0.05), spleen and thymus index, the level of macrophages phosphatase activity (ACP) and mRNA expression of IL-2, IL-4and INF-a were extremely increased(.P<0.01), the level of IgM and daily gain were significantly increased(P<0.05).(2) In perillaldehyde group, the level of macrophages phosphatase activity (ACP) and mRNA expression of IL-2and INF-a were extremely increased(P<0.01), daily gain and blood sugar was increased(.P<0.05).(3)In limonene group, the spleen index and mRNA expression of IL-4and INF-awere extremely increased(P<0.01).
