Grafting techniques were the effective means to overcome the continuous cropping obstacles which could lead to soils environmental degradation, soil-borne diseases, yields and quality declining in muskmelon. The seriously excessive vegetative growth, which were usually occurred by conventional grafting in low light, high temperature and humidity environmental before the grafting survival, would been prevented by rootstocks root-cut-grafting. This study research the effects of rootstocks root-cut-grafting on growth and yield of muskmelon by screening the different soil temperature, growth regulators and rootstocks of variety species and seedling age. The results showed that,(1) Three suitable rootstocks named Z5, Z8and Z9, whose optimum temperature for regeneration root growth was20℃, were screened from the nine rootstocks. However, different rootstocks had their different root cutting age, in our study, root cutting age of Z5and Z8were4d after cotyledons flatted, and Z9was6d after cotyledons flatted. The regenerative capacities of rootstocks after root-cutting, which were treated with IB A:NAA=1:1, were stronger then the one which was treated alone with IBA or NAA, in addition, the best treatment of Z5was3000times liquid of IBA:NAA=1:1by root dipping, so did the Z8with4000times liquid of IBA:NAA=1:1by leaf spraying and Z9with3000times liquid of IBA:NAA=1:1by leaf spraying.(2) The root activity of rootstocks regenerated roots was positive correlated to the root surface area, and root volume. The root activity would raise gradually and root surface area, root volume would increase rapidly as the growth of regenerated roots.(3)The root activity, root surface area and root volume of Z5, Z8and Z9rootstocks regenerated roots were increased as the temperature and root cutting age increased, however, they could been decreased when the temperature and root cutting age continued to increase. And the root activity, root surface area and root volume of rootstocks regenerated roots would reached their maximum value when cotyledons of Z5and Z8were flatted for4d, and cotyledons of Z9was flatted for6d in20℃.(4) The root activity, root surface area, root volume and catalase activity of rootstocks regenerated roots were increased after IBA or NAA or IBA:NAA=1:1treating, as did the IAA oxidase, Ca2+-ATPase and Mg2-ATPase activity in previous period of root cutting. However, CAT, Ca2+-ATPase and Mg2-ATPase activity could been first increased and then decreased during the root regeneration, when IAA oxidase activity could been first decreased and then increased.(5) Compared with the conventional grafting, the ZT, IAA and GA content of rootstocks root-cut-grafting could increase. The ZT and IAA contents were first decreased and then increased after root cutting, however, GA contents were increased after root cutting.(6) Compared with the conventional grafting, the grafting survival rate, root activity, root surface area, root volume and yield of rootstocks root-cut-grafting could increased effectively. In our study, grafting survival rate of Z5and Z8rootstocks with root-cut-grafting were obviously higher than the others. The soluble sugar and vitamin C contents of muskmelon by conventional grafting or root-cut-grafting were significantly higher than own-rooted muskmelon, in addition, soluble sugar and vitamin C contents of Z9and Z8rootstocks with root-cut-grafting were obviously higher than the one with conventional grafting, nevertheless, soluble sugar and vitamin C contents of Z5rootstocks with root-cut-grafting were significantly lower than the one with conventional grafting.
