In this study, seedling top leaves, leaves callus, seeds with breaking the dormancy of Vaccinium uliginosum were as the test materials, induced polypioidy with colchicine and oryzalin; preliminarily ldentificated the induced polypioidy plants which lays the foundation on the innovation, protection, and utilization of germplasm resources, the results were as follows:1. The seeds inoculated in Read culture medium with gibberellin1.0mg·L-1, after7days dark culture and then light culture, the germination rate was33.3%, seeds which treated with light culture do not germinate.2. The seeds stored at low temperature30days, with400mg·L-1gibberellin soaking24hours, under the condition of low temperature which stratification30days, what can be effectively on breaking seeds dormancy and the germination has the best effect, the germination potential and germination rate of the average value were respectively46.67%and95.56%.3. Compared to the induced polypioidy survival effect on the Vaccinium uliginosum. oryzalin induced better than colchicine, it has low toxicity on plants, and induced agent added to the medium which is better than the immersion method, leaves callus which was as induced test material that is better than top leaves and the seeds of breaking dormancy.4.The leaves callus of Vaccinium uliginosum were as induced materials, preliminary identification found that stoma density of variation which is lower than control plants, stomata guard cell size of variation which is bigger than control plants, the variation which numbers of stomata guard chloroplast cell were between8-11may be polyploidy.5. Colchicines added to the medium which concentration was400mg·L-1, the chloroplast variation rate of guard chloroplast cell of plant regeneration leaves when exposure21d was83.3%;oryzalin added to the medium which concentration was40mg·L-1, the variation rate was highest when exposured14d, it could reach44.7%.
